摘要
目的 :体外观察染料木黄酮 (genistein)对颅盖骨骨吸收活性的作用。方法 :分离乳鼠颅盖骨 ,分别加入 0、10 -8和 10 -6mol/L的染料木黄酮 ,并以 10 -9mol/L的 17β -雌二醇 (17βE2 )为对照 ,检测培养液上清中Ca2 + 含量和酸性磷酸酶活性 ,以评价染料木黄酮对颅盖骨骨吸收活性的作用。结果 :10 -8和 10 -6mol/L染料木黄酮使培养液上清中Ca2 + 含量下降 ,吸收指数 <1.0。 10 -9mol/L 17βE2 的吸收指数介于 10 -8与 10 -6mol/L染料木黄酮之间。酸性磷酸酶活性在各组之间无差异。结论
AIM:To investigate the effect of genistein, a soybean-derived isoflavone, on the bone resorption in the cultured calvarium.METHODS: The calvaria were obtained from 5d-old rats and cultured in the medium of 0,10 -8 and 10 -6mol/L of genistein respectively,with 10 -9mol/L 17β-estradiol(17βE 2) as control.The content of acid phosphatase (ACP) and Ca 2+ were tested to evaluate the bone resorption of mice calvarium. RESULTS:The content of Ca 2+ decreased in the concentration of 10 -8 and 10 -6mol/L of genistein.The index of bone resorption of 10 -9mol/L 17βE 2 lied between 10 -8 and 10 -6mol/L of genistein. There were no difference in the contents of ACP among all groups.CONCLUSION:Genistein can regress the bone resorption in the cultured calvarium.
