摘要
目的 :探讨上皮钙粘附蛋白 (E -CD)、α -连环蛋白 (α -CA)及 β -连环蛋白(β -CA) 表达与大肠癌临床病理学因素的关系 ;方法 :应用免疫组织化学ABC法检测 76例大肠癌组织及临近正常粘膜均有E -CD、α -CA和β -CA的表达 ;结果 :正常大肠粘膜均有E -CD、α-CA和 β-CA的强烈表达。 48.7%大肠癌组织E -CD表达减弱 ,E -CD表达减弱与大肠癌的分化程度相关 (P <0 .0 1 ) ,但与大肠癌的Dukes分期及淋巴结转移无关 (P >0 .0 5)。 76例大肠癌中 42例 (55.3 % )α -CA和 3 2例 (42 .1 % ) β -CA表达减弱 ,并且在分化较差、Dukes分期较晚及伴有淋巴结转移的大肠癌组织中α-CA和β-CA表达减弱更加明显 (P <0 .0 1 )。E -CD、α -CA和 β -CA中至少一项表达减弱者 54例(71 .1 % ) ,且与大肠癌分化程度、Dukes分期及淋巴结转移的关系更为密切 (P <0 .0 1 ) ;结论 :结合分析E -CD、α -CA和β -CA的表达特征有助于大肠癌生物学行为及预后的判断。
Objective:To investigate the relationship between the expression of E-cadherin(E-CD), α -catenin(α-CA) and β-catenin (β-CA) and clinical pathological parameters in colorectal cancer.Methods: Avidin-biotin complex method was used to study the expression of E-CD, α-CA and β-CA in different tissues from 76 cases of colorectal cancer, including the primary tumor and normal colorectal mucosa nearby the tumor. Results: The normal colorectal mucosa strongly expressed E-CD, α-CA and β-CA. Reduced E-CD expression was found in 48.7% colorectal cancer, which was associated with tumor differentiation colorectal cancer (P<0.01). There was no significant association between E-CD expression and Dukes stage and lymph node metastasis (P>0.05). The expression of α-CA and β-CA decreased in 55.3% and 42.1% colorectal cancer, respectively, and reduced expression of α-CA and β-CA was found more frequently in poorly differentiated tumors, in tumors with Dukes stage C or D and in tumors with lymph node metastasis (P<0.01). The expression of at least one of the investigated molecules was reduced in 71.1% colorectal cancer, which was more significantly associated with tumor differentiation, Dukes stage and lymph node metastasis (P<0.01). Conclusion: The combined analysis of E-CD , α-CA and β-CA expression will be helpful in predicting the biological behavior and prognosis of colorectal cancer.
