吗啡对大鼠催乳素基因表达的影响

摘要 目的 :研究吗啡依赖及戒断对大鼠垂体催乳素 (PRL)基因表达的影响。方法 :核酸分子杂交及放射免疫分析。结果 :吗啡依赖组及戒断组大鼠垂体的PRLmRNA含量降低 ,戒断 2组与对照组之间差异有显著性 (P <0 0 5 ) ;吗啡依赖组及戒断组血清PRL水平均高于对照组 ,后者与对照组之间差异有显著性 (P <0 0 5 )。结论 :吗啡依赖及戒断显著影响大鼠垂体PRL的基因表达 。 Objective: To investigate the gene expression of prolactin in hypophysis of experimental rats which were exposed to morphine dependence and withdrawal. Methods: Prolactin concentration of morphine dependent and withdrawal rats were determined by radioimmunoassay. The prolactin gene expression was investigated by dot and Northern blot hybridization. Results: Compared with the control, the gene expression both in morphine dependence group and morphine withdrawal group decreased. There was significant difference between morphine withdrawal group two and the control group (P<0.05). The concentration of prolactin in the serum of morphine dependence group and morphine withdrawal group were higher than that in control group, showing significant difference (P<0.05). Conclusion: By decreasing the transcription and stimulating the secretion directly, morphine dependence and withdrawal can affect the gene expression of prolactin in hypophysis significantly.
