摘要 目的 :以盐酸吗啡为阳性对照药 ,观察雷米芬太尼对动物的镇痛强度及时效关系。方法 :静脉注射雷米芬太尼后用热板法、夹尾法、甲醛法和甩尾法评价雷米芬太尼对大、小鼠的镇痛强度 ;用大鼠甩尾法测定镇痛时效曲线。结果 :小鼠热板法镇痛ED50 值及 95 %可信限为 0 .73(0 .6 4 - 0 .84 )mg·kg- 1 ;小鼠夹尾法ED50 值及 95 %可信限为 0 .19(0 .12 - 0 .31)mg·kg- 1 ;大鼠甲醛法镇痛ED50 值及 95 %可信限为 2 .70 (1.15 - 6 .34) μg·kg- 1 ;大鼠甩尾法镇痛ED50 值及 95 %可信限为 5 .2 1(2 .11- 12 .85 ) μg·kg- 1 ;雷米芬太尼在iv后 1min镇痛作用最强 (P <0 .0 1) ,6min后作用减弱 ,12min后作用基本消失。结论 :雷米芬太尼的镇痛作用比吗啡强 ,似有种属差异 ,小鼠相对不敏感。作用时间短暂 ,提示雷米芬太尼临床比较适合静脉点滴做长时间的外科手术及术后的镇静 。 Objective: To observe the analgesic effect and time-effect relation of Remifentanil on animal, and compare with morphine hydrochloride. Methods: The analgesic intensity of Remifentanil on mice and rat were evaluated after intravenous injection using hot-plat and clam-tail methods in mice and formaldehyde and swing-tail methods in rat. Analgesic time-effect curve of Remifentanil were determined by swing-tail method in rat. Results: ED 50 and 95 percent confidence limits of analgesic effect of Remifentanil on mice were 0.73(0.64-0.84) mg·kg -1 and 0.19(0.12-0.31)mg·kg -1, respectively in hot-plat and clam-tail methods;ED50 and 95 percent of analgesic effect of Remifentanil on rat were 2.70(1.15-6.34)μg·kg -1 and 5.21( 2.11-12.85)μg·kg -1, respectively in formaldehyde and swing-tail methods. The analgesic action of Remifentanil was strongest at 1 minute after intravenous injection. The action was weakened after 6 minutes,and disappeared after 12 minutes. Conclusion: Analgesic effect of Remifentanil was stronger than morphine hydrochloride. This probably has ethnic difference. Mice is relatively insensitive. Analgesic effect of Remifentanil is very short. It is suggested that Remifentanil be suitable by intravenous instillation for surgical operation, sedation after operation and can be used as adjuvant for anesthesia.
