摘要
①目的 探讨二氧化矽 (SiO2 )对人肺泡巨噬细胞基质金属蛋白 (MMP - 9)表达的影响。②方法 收集矽肺患者肺泡巨噬细胞 (AM) ,在体外以 50 μg/mlSiO2 刺激、LPS或无血清培养基培养 2、6、1 2、1 8、2 4、36h ,用免疫细胞化学的方法检测AM中MMP - 9的表达。③结果 经SiO2 刺激的AM中MMP - 9表达明显上调 ,且随刺激时间不同表达量也不同 ,于 1 8h达高峰。不同时间的表达量与同期对照组AM相比差异显著 (P <0 .0 1 )。④结论 SiO2 可诱导AM高表达MMP - 9,提示AM在损伤早期通过产生MMP - 9而降解基底膜 ,可能与包括成纤维细胞在内的各种细胞的迁移有关。
Objective To study the effect of SiO 2on the expression of MMP-9 in human alveolar macrophages. Methods AM were collected from a silicotic by BAL and exposed to SiO 2 for different time, then the immunocytochemical method was used to detect the level in the expression of MMP-9 in AM. At light microscopical level, the staining sample were analysed quantitatively by automatic image analytical system. Results The patient AM exposed to silica (50μg/ml) could express MMP-9 and at 18h the quantities of MMP-9 expressed by AM reached the peak. After that, the expressed quantities reduced markedly. Compared with control, the quantities of MMP-9 expressed in AM exposed silica ware much more. Conclusion SiO 2 could induce the expression of MMP-9 in AM. It's inferred that MMP-9 produced by AM at early stage of lung injury has effect on immigrate various cells including FB into the injured region by degrading the base membrane.
