摘要
观测核因子 κB(nuclearfactorkappaB ,NF κB)调控脂多糖 (lipopolysaccharide ,LPS)激活的中性粒细胞 (polymor phonuclearneutrophil,PMN)凋亡的下游相关基因。将培养的人静脉血PMN分为对照 (control)组、LPS(10 0 μg/L)组、gliotoxin +LPS组及PD0 980 5 9+LPS组 ,后两组先用gliotoxin(10mg/L)或PD0 980 5 9(10 0 μmol/L)培养 30min ,后加入LPS(10 0 μg/L)继续培养 6 0min。收集细胞 ,提取总RNA。采用细胞凋亡相关蛋白及应激反应蛋白类基因芯片进行检测。结果显示 ,control组与LPS组有 8条差异基因 ,3条上调 ,5条下调 ;gliotoxin +LPS组与LPS组有 10条差异基因 ,7条上调 ,3条下调 ;PD0 980 5 9+LPS组与LPS组有 8条差异基因 ,5条上调 ,3条下调。综合分析发现Defenderagainstcelldeath 1和X linkedinhibitorofapoptosisprotein是NF κB调控PMN凋亡的下游相关基因。Ionizingradiationresistancecon ferringprotein在NF
To investigate the downstream genes of lipopolysaccharide(LPS) stimulated PMN apoptosis regulated by NF-κB, the cultured human venous blood PMNs of four groups as follows were studied: ①control group incubated with sterile saline (10mL/L), ②LPS (100μg/L) group, ③gliotoxin+LPS group and ④PD098059+LPS group. The last two groups were pretreated with gliotoxin (10mg/L) or PD098059 (100μmol/L) 30min before LPS challenge PMNs were harvested one hour later for total RNA extraction. The downstream genes of NF-κB to regulate LPS-stimulated PMN apoptosis were explored with the gene chip representing apoptosis and stimulation-related proteins. The results showed that three genes were up-regulated and five genes down-regulated between control group and LPS group, seven genes up-regulated and three down-regulated between gliotoxin+LPS group and LPS group, five genes up-regulated and three down-regulated between PD098059+LPS group and LPS group. These results indicated that the genes named as defender against cell death 1 and X-linked inhibitor of apoptosis protein were downstream genes of NF-κB regulating LPS-stimulated PMN apoptosis. Further studies were needed to prove that ionizing radiation resistance conferring protein is regulated by NF-κB as a downstream gene.
