施CO_2时培养基糖浓度对文心兰试管苗生长的影响 被引量:10

摘要 探讨了培养室内施CO2条件下,培养基中不同蔗糖浓度(30、20、10、5、2.5和0 g·L-1)对文心兰试管苗生长的影响。结果表明:无糖培养基中的试管苗可以以光合自养的方式正常生长,但生长速度略低;提高培养基糖浓度可以显著促进试管苗的生长,30 g·L-1(常规培养条件的糖浓度)与20 g·L-1处理相比,株高、叶长、叶幅、根数、总鲜样质量和总干样质量等主要生长指标无明显差异,说明文心兰试管苗在施CO2培养时,培养基中的糖仍是必要的,但可以适当减少用量。 Oncidiwn aloha 'Iwanga' in vitro shoots were used as plant materials for experiments to study the effect of different sucrose concentrations (30, 20, 10, 5, 2.5, 0 g·L-1) in the culture medium on the growth of the in vitro shoots under CO2 enrichment conditions. The results showed that the in vitro shoots had photoau-totrophic capability. Under CO2 enrichment conditions, in vitro shoots being cultivated in the sugar-free culture medium could grow normally by photoautotrophic capability, but a bit slowly. Increasing the sucrose concentration in the culture medium could significantly promote the growth of the in vitro shoots of Oncidiwn. Under CO2 enrichment condition, in vitro shoots grew better in the sugar culture medium than in the sugar-free culture medium. Compared with the sucrose concentration (30 g·L-1) under normal culture conditions, when the sucrose concentration dropped to 20 g·L-1, there was not significant difference between the two concentration treatments in the main growth indices of the in vitro shoots including plant height, length of leaves, range of leaves, number of roots, total fresh mass and dry matter mass. The above results indicate that under CO2 enrichment culture conditions, the existence of sucrose in the culture medium is still necessary for Oncidium, but compared with normal culture methods, it can partly reduce the amount of sucrose used in the culture medium.
