摘要
以华阳三号(HY)大白菜和苏州青(SZQ)小白菜下胚轴为外植体,MS为基本培养基,将TDZ与NAA配置不同激素浓度组合,观察TDZ对下胚轴不定芽再生的影响,并对各组合的培养效果进行了比较。结果表明,TDZ能较好地促进大白菜下胚轴不定芽再生,在MS+0.5 mg/L TDZ+0.5 mg/L NAA的培养条件下,HY的不定芽再生频率达到45%,且再生周期短,芽点多,再生芽正常,可以满足植物遗传转化需要。TDZ很难诱导SZQ小白菜下胚轴不定芽再生,只能诱导产生大量愈伤组织。
Effects of TDZ on regeneration of hypocotyl explants from Chinese cabbage (Brassica campestris L. ssp. pekinensis cv. HY) and pak-choi ( B. campestris L. ssp. chinensis cv. SZQ) were investigated and compared on MS basic medium supplemented with different concentrations of TDZ and NAA. The results showed that TDZ was effective for shoot regeneration from hypocotyl explants of HY and its regeneration frequency was up to 45% in the medium of MS + 0. 5 mg/L TDZ+ 0. 5 mg/L NAA, the shoot regeneration was more efficient and rapid on this medium suitable for plant genetic transformation. It was difficult to induce shoot buds from hypocotyls of pak-choi SZQ in all of these media containing TDZ while there were many calli developed.
