脑胶质瘤细胞SHG-44的热、放射敏感性及Fas/FasL表达 被引量:1

摘要 观测加热联合放射对SHG-44细胞的影响及相关基因Fas/FasL的表达。采用细胞集落法及流或细胞术观察加热对脑胶质瘤细胞SHG-44放射后集落形成的影响和凋亡的变化,并用RT-PCR法检测Fas/FasL表达。结果表明,放射+42℃加热(加热时间40min)较单纯放射(2Gy)的细胞集落明显减少,热增强比(TER)为1.95;空白对照组、单纯放射组、放射+加热组凋亡比率分别为:20.2、29.3、59.1;Fas在3个实验组中均表达,但FasL仅在单纯放射组、放射+加热组中表达。热能明显增加脑胶质瘤细胞SHG-44的放射敏感性,也表明凋亡与Fas/FasL有关。 The effects of hyperthermia and radiation on human glioma SHG-44 cells and the alteration of Fas/FasL expression in the treated cells were investigated. The sensitivity of SHG-44 cells to hyperthermia and radiation was measured by colony forming assay and flow cytometry (FCM). The expression of Fas/FasL was detected by RT-PCR. By hyperthermia ( 42℃, 40min) combined irradiation, the colony forming ratio was reduced in a more effective way than by just the irradiation. The TER was 1.95. The apoptosis ratios were 20.2, 29.3, 59.1%, respectively, for the control group , the just-irradiation group and the hyperthermia combined radiation group. The expression of Fas was found in the three groups, but the expression of FasL could be found only in the two treatment groups. Hyperthermia could boost the effectiveness of radiation against glioma cells significantly. This also indicates some relationships between apoptosis and Fas/FasL.
