电离辐射诱导BALB/c小鼠骨髓细胞基因表达谱的研究 被引量:8

摘要 利用基因芯片技术研究了辐射后小鼠骨髓细胞基因表达谱的变化。结果发现在芯片上8079个基因中,有660个基因的表达有明显差异,其中354个基因的表达量增高,306个基因的表达量下降。在差异基因中功能或部分功能已知基因有222个,对其中差异2.5倍以上基因进行分析,发现表达差异基因涉及细胞凋亡、细胞周期、信号转导、免疫与应激、核酸合成、转运蛋白与通道蛋白等多个方面,其中以信号转导和免疫应激相关基因所占比例较多,反映出辐射对细胞损伤的多靶点、多层次及多通路的特点,深入研究这些辐射相关基因在造血辐射损伤中的作用,可为放射病治疗、肿瘤放疗提供新的靶点和方法。 Microarrays except for cDNA expression arrays was used to profile changes in gene expression of bone marrow cell after exposed BALB/c to ionizing radiation (IR). The RNA from bone marrow cells was subjected to cDNA analysis on a microarray of 8079 known genes and ESTs.Data analysis showed that 660 genes changed at least 2-fold.among the 660 genes, 354 genes showed up-regulation in the radiated BALB/c, whereas 306 genes were down-regulated. 222 of the 660 genes are function known or inferred genes, which fell into categories of signal transduction, immunity or defense response, nucleic acid correlated, apoptosis, cell cycle, transport, metabolism etc. It is suggested that a variety of regulatory pathways in hematopoietic cells were affected by irradiation.
