植物青枯病高效生防菌株PBW1培养基优化和生长特性研究

摘要 为了更好地对植物青枯病高效生防菌株 PBW1进行培养 ,采用正交试验的方法研究了生防菌株 PBW1的最佳培养基组成 ,结果为葡萄糖 2 5 g/L、淀粉 2 0 g/L、蛋白粉 4g/L、酵母粉 5 g/L;同时对 5 0 0 m L摇瓶培养过程中菌体生长、还原糖、总糖及 p H变化特征进行了研究。此外 ,还在 6L全自动发酵罐中对 PBW1培养过程中的菌体生长、还原糖、氨基氮、p H和 DO的变化特征进行了研究 ,培养 48h的菌体浓度达 4.8× 1 0 12 CFU/m L。研究结果为 The culture medium of bio-controlling strain PBW1 was studied with single-factors and orthogonal tests, and the bio-controlling strain PBW1 of plant bacterial wilt disease was cultured well. The optimum culture medium ingredients of PBW1 in 500 mL flask were obtained as follows: glucose 25 g/L, starch 20 g/L, protein powder 4 g/L, yeast powder 5 g/L. With the optimum culture technology, the characteristics of cell growth, consumption of reduced sugar and total sugar as well as pH were also studied. Besides, the characteristics of cell growth, consumption of reduced sugar and NH_2-N, pH as well as DO were investigated in the process of PBW1 culture within a 6 L automatic fermentor and cell concentration in the medium reached 4.8×10~ 12 CFU/mL when it had been cultivated for 48 hours. The above results set a good basis for large-scale culture technology and the scale-up of PBW1 culture process.
