吡格列酮对实验性大鼠糖脂代谢紊乱的影响 被引量:2

摘要 [目的 ]研究胰岛素增敏剂吡格列酮对糖脂代谢紊乱的影响。 [方法 ]以实验性糖脂代谢紊乱大鼠为模型。SD大鼠 30只随机分为正常对照组、高脂模型组和给药组。正常对照组喂饲普通饮食 ,给药组在高脂饮食 4周后予吡格列酮 1 0mg·kg- 1·d- 1灌胃 ,至第 9周末处死各组大鼠。 [结果 ]9周模型组出现明显腹型肥胖和糖脂代谢紊乱 ,给药组与模型组相比肝湿重、肾周脂肪重、Lee’s指数、肝指数、脂比重明显降低 ,分别为 (9.0 6± 1 .2 0 ) g比 (1 3.2 3± 1 .88) g、(2 .1 2± 0 .5 5 ) g比(4 .2 4± 1 .1 1 ) g、(2 80 .1 1± 7.5 9)比 (2 95 .89± 9.36 )、(3.0 2± 0 .31 ) %比 (4 .0 9± 0 .36 ) %、(0 .36±0 .1 3) %比 (0 .6 7± 0 .1 5 ) % (P <0 .0 1 ) ;给药组血清TC、LDL -L、FFA均有显著的降低 ,分别为(1 .99± 0 .5 1 )比 (3.2 8± 1 .0 8)mmol/L、(1 .1 6± 0 .37)比 (2 .1 7± 0 .82 )mmol/L、(374 .6 4± 2 5 .89)比 (4 6 3.6 3± 2 2 .39) μmol/L(P <0 .0 1 )。 [结论 To study the effect of Pioglitazone on experimental lipid metabolism disorder model of rats induced by high-fat diet. 30 SD rats were randomly allocated into 3 groups;Model group ( n =10) fed with high fat diet,and treated group ( n =10) fed with high fat diet for 4 weeks added pioglitazone (10 mg·kg -1 ·d -1 ),and normal group ( n =10) fed with standard diet for 9 weeks and sacrificed. After 9 weeks, Model rats showed to be abdomen-form obesity and developed glucose -lipid metabolism disorder;Compeared with the model group ,liver moist weight? perirenal fat weight?Lee,s exponent?liver exponent and fat density of the treated group were (9.06±1.20)g vs(13.23±1.88)g?(2.12±0.55)g vs(4.24±1.11)g?(280.11±7.59)vs(295.89±9.36)?(3.02±0.31)% vs(4.09±0.36)%?(0.36±0.13)% vs(0.67± 0.15) %( P <0.01);The levels of TC?LDL-L?FFA were(1.99±0.51)vs(3.28±1.08)mmol/L?(1.16± 0.37) vs(2.17±0.82)mmol/L and(374.64±25.89)vs(463.63±22.39)μmol/L( P <0.01). [Conclution] Pioglitazone is effective in the treatment of glucose -lipid metabolism disorder rats induced with high-fat diet.
