摘要
目的:研究牛脑提取液对脊髓损伤修复及再生的影响。方法:用SD大鼠40只,过半切除长约0.5cm的左下胸段脊髓,肌基膜管(MBL)桥接脊髓缺损。局部用药组将MBL浸泡于牛脑提取液(ECB)中,全身用药组腹腔注射ECB,生理盐水作对照。结果:实验组动物行为学测试指标和斜板试验斜板实验(IP)分数均明显优于对照组;实验组术后6周可测到体感诱发电位(SEP)波形,而对照组未测到;实验组MBL与脊髓融和,有大量神经纤维通过交界处,近端脊髓前角区可见到较多标记的运动神经元,腰部脊神经节细胞结构完整,再生神经纤维髓鞘较厚,轴突直径较粗,均优于对照组;图像分析显示实验组再生有髓神经纤维数、轴突直径和脊神经节细胞数均优于对照组。结论:局部和全身应用ECB对脊髓再生均有明显促进作用,尤其对脊髓运动神经元的再生也有显著促进作用。
AIM:To study the effect of extraction of calf brain (ECB) on regeneration and repair of spinal cord injury. METHODS:Forty SD rats were used in this test, 0.5 cm left interior spinal cord was half past removed, and muscle basal lamina (MBL) was bridged the gap.Extraction of calf brain was either immersed locally or administrated systematically by celiac injection. Normal saline was used for control groups. RESULTS:It showed that behavior survey and inclined plane test were better in experimental groups than in control groups;six weeks after operation somato sensory evoked potential(SEP)was measured in experimental groups, there was no SEP in control groups; the spinal cord MBL interface healed well,a large amount of regenerated spinal axons grew across the bridge.Many labeled motor neuron were found at anterior horn of proximal spinal cord,lumbar spinal cord nerve ganglion cells had integrate appearance.In these axons of regenerative nerve fibers, there were thicker myelin and wider in diameter,and these phenomena were better than the control groups; image analysis showed that the number of nerve fibers, the diameter of axons and the number of lumbar spinal cord ganglion cells were better in experimental groups than in control groups. CONCLUSION:ECB obviously promote the regeneration of spinal cord with both local and systematically administration especially for the regeneration of spinal cord motor neuron.
