丹参水溶性主成分丹参素和阿魏酸合用的药动学相互作用 被引量:10

摘要 目的研究丹参水溶性主成分丹参素和川芎水溶性主成分阿魏酸合用时的药动学相互作用。方法6只♂健康家兔,采用随机交叉与自身对照分组,分别进行单用丹参水溶性主成分丹参素和阿魏酸及同时合用丹参素和阿魏酸时的主要药动学参数比较。采用反相高效液相色谱法测定家兔血浆中丹参素及阿魏酸的浓度。结果静脉给药后,丹参素和阿魏酸在家兔体内符合二室开放模型,在单用和复合给药时的分布半衰期(T12α),消除半衰期(T21β),总体消除率(Cls),药-时曲线下面积(AUC)等主要药动学参数均无统计学显著性差异。结论在给药剂量下,丹参素与阿魏酸合用时,没有表现出明显的药动学相互作用。 Aim To investigate the pharmacokinetic interaction between ferulic acid and danshensu, the hydrophilic principal components of danshen (Salvia miltiorrhiza Bung) in rabbits after administrating a single intravenous dose. Methods The rabbits were divided into three groups with an open randomized crossover and self-control design. The concentrations of ferulic acid and danshensu in rabbit plasma were determined with RP-HPLC method. The main pharmacokinetic parameters of ferulic acid, danshensu and the co-administration of ferulic acid and danshensu were investigated respectively. The data of concentration-time curve of danshensu and ferulic acid were calculated by 3P87 program. Results The plasma drug concentration-time courses of ferulic acid and danshensu in rabbits were both fitted to a two-compartment open model. There were no significant differences in the main pharmacokinetic parameters such as T12α, T12β, Cls and AUC between the single injection group and the combination injection group. Conclusion There was no evidence of pharmacokinetic interaction between the hydrophilic principal components of danshen and ferulic acid after administrating intravenous combination dose in rabbits. The results provided a useful index for clinical practice.
