夏枯草中迷迭香酸含量测定及UPLC特征图谱研究 被引量:4

摘要 目的通过夏枯草中迷迭香酸的高效液相色谱(HPLC)含量测定及超高效液相色谱(UPLC)特征图谱研究,为夏枯草质量评价提供依据。方法采用HPLC法对16批夏枯草的迷迭香酸进行测定;采用UPLC建立夏枯草特征图谱,以Waters Acquity UPLC?HSS T3(2.1 mm×100 mm,1.8μm)为色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)作为流动相进行梯度洗脱:0~1 min,10%A;1~3 min,10%~15%A;3~6 min,15%A;6~8 min,15%~23%A;8~14 min,23%A;14~18 min,23%~30%A;18~22 min,30%~38%A;22~24 min,38%~90%A;24~26 min,90%A;26~28 min,90%~10%A;柱温:30℃,流速:0.4 mL·min-1,检测波长:280 nm。结果夏枯草的成熟度和迷迭香酸含量有相关关系;特征图谱确定其中6个共有峰,相似度在0.95以上。结论所建立的方法快速、准确、可靠,可以为夏枯草药材的质量控制提供依据。 OBJECTIVE To establish the characteristic chromatogram of herbs Prunella vulgaris by UPLC,and determine the rosmarinic acid by HPLC,so as to provide basis for controlling its quality.METHODS The contents of rosmarinic acid of 16 samples were determined by HPLC.The UPLC characteristic chromatogram was performed on a Waters Acquity UPLC?HSS T3(2.1 mm×100 mm,1.8μm)column,with acetonitrile as mobile phase A and 0.1%aqueous phosphoric acid solution as B,with gradient elute:0-1 min,10%A;1-3 min,10%-15%A;3-6 min,15%A;6-8 min,15%-23%A;8-14 min,23%A;14-18 min,23%-30%A;18-22 min,30%-38%A;22-24 min,38%-90%A;24-26 min,90%A;26-28 min,90%-10%A.Column temperature was set at 30℃,the flow rate was 0.4 mL·min-1,and the detective UV wavelength was 280 nm.RESULTS The content of rosmarinic acid was related to the maturity of herbs Prunella vulgaris.The characteristic chromatogram retained 6 common peaks,and the similarities were all above 0.95.CONCLUSION The established methods are rapid,accurate and repeatable,which can be used to quality control of herbs Prunella vulgaris.
