COX-2 mRNA在膝骨关节炎病变过程中的作用研究

Study on the Role of COX-2 mRNA in the Pathological Process of Knee Osteoarthritis

目的探讨COX-2 mRNA在膝骨关节炎(KOA)病变过程中的作用。方法 76例膝骨关节炎患者(研究组)根据病变进展分为初期、中期、晚期三组,选取15例非膝骨关节炎患者为对照组,比较各组的COX-2 m RNA以及COX-2蛋白表达水平。结果研究组、初期组、中期组、晚期组的成纤维样滑膜细胞中COX-2 mRNA及蛋白表达灰度值显著高于对照组(P<0.05);初期组的成纤维样滑膜细胞中COX-2 mRNA及蛋白表达灰度值显著高于中期组、晚期组(P <0.05)。结论 COX-2mRNA在膝骨关节炎患者病变初期表达水平较高,检测COX-2 m RNA表达水平有助于为膝骨关节炎患者的诊治及预后提供指导。

Objective To investigate the role of COX-2 m RNA in the pathological process of knee osteoarthritis(KOA).Methods A total of 76 cases of KOA patients(study group)were divided into the early stage group,middle stage group and late stage group according to the pathological progress of KOA.15 cases of patients with non-knee osteoarthritis were selected as control group.The expression levels of COX-2 m RNA and COX-2 protein of each group were compared.Results The gray values of COX-2 m RNA and protein expression in fibroblast synovioid cells of the study group,early stage group,middle stage group and late stage group were all significantly higher than those of the control group(P<0.05).The gray values of COX-2 m RNA and protein expression in fibroblast synovioid cells of the early stage group were significantly higher than those of the middle stage group and late stage group(P<0.05).Conclusions COX-2 m RNA expression is higher in patients with knee osteoarthritis at the early stage,and the detection of COX-2 m RNA expression level is helpful to provide guidance for the diagnosis,treatment and prognosis of patients with knee osteoarthritis.
