疱疹病毒感染后HEp-2细胞MDH变化的初步观察

摘要 HSV-Ⅰ感染 HEp-2细胞后,胞浆及线粒体苹果酸脱氢酶(MDH)活力的升高分别以12h、44h 以及4h、16h、28h、32h 最明显:HSV-Ⅱ感染后,胞浆及线粒体 MDH 活力的升高分别以24h、48h 及4h、8h、16h、32h 最明显.用各型抗血清及紫外线灭活后,胞浆及线粒体 MDH 活力明显下降.方差分析及 q 检验表明这些变化具有高度显著性.MDH 酶谱分析可看出病毒感染主要与胞浆内分子量(MW)为67000和525000道尔顿(d)和线粒体内 MW36000d 的带有关.并对泪液和脑脊液 MDH 活性测定的临床应用进行了讨论. In HEp-2 Cells infected by HSV-Ⅰ,the activity of cytosol and mitochondrial MDH increased most evidently at the 12,the 44,the 4,the 16,the 28 and 32nd hour post infection.After HSV-Ⅱ infection,the MDH activity raised most remarkably at the 24,the 48,the 4,the 8,the 16 and 32nd hour post infection.When the samples were treated with corresponding antiserum and inactivated by UV light,their MDH activities decreased evidently. Statistical Analysis and the q test indicated that these changes were significant.Analysis of MDH isozyme patterns revealed that about 6700 and 525000 dalton bands in cytosol MDH and about 3600 dalton band in mitochondrial MDH were related with virus infection.The signifi- cance of measurement of MDH activity in tear and cerebrospinal fluid in clinies was discussed.
