金银花组织培养外植体灭菌方法的研究

Study on Sterilization Method of Tissue Culture Explants of Lonicera japonica

近年来金银花的需求量不断上升,价格呈现攀高的趋势,金银花种苗价格也不断升高。大田生产中大多通过扦插和分根等方式进行繁殖,但品质却不断下降。如何快速得到大批量、高品质和低成本的种苗,组织培养是最好的方式之一。本实验通过使用不同浓度的碱化乙醇和0.1%的氯化汞进行灭菌处理,通过试验发现经质量分数为0.1%氯化汞7 min处理后灭菌效果最佳,成活率能达到88.3%,污染率低至8.3%。同时,经过初步清洗处理后接种于含0.1%纳米银分子的培养基中的外植体更是达到了98.3%的灭菌率,成活率高达96.7%。这种低成本高效的灭菌方式有利于脱毒苗的大批量生产,为大田生产提供了便利条件。

In recent years,the demand of honeysuckle(Lonicerae Flos)has been increasing,and the price of seedlings has been rising.In the field,most of them are propagated by cuttage planting and root splitting,but the quality is decreasing.Tissue culture is one of the best ways to get mass,high quality and low cost seedlings quickly.In this experiment,different concentrations of alkalized ethanol and 0.1%mercuric chloride were used to sterilize.It was found that the best sterilization effect was obtained after 7 minutes of 0.1%mercuric chloride treatment in single factor experiment.The survival rate was 88.3%and the pollution rate was 8.3%.At the same time,the explants inoculated in the medium containing 0.1%silver nanoparticles after simple preliminary cleaning treatment reached 98.3%sterilization rate,and the survival rate was as high as 96.7%.This low cost and high efficiency sterilization method is conducive for mass production of virus-free seedlings,and provides convenient conditions for field production.
