尼克酸治疗恶性黑素瘤的相关机制研究

摘要 目的探讨尼克酸在恶性黑素瘤的治疗前景及相关分子机制方法通过Transwell体外迁移实验,观察尼克酸对B16-F10黑色素瘤细胞迁移是否有抑制作用,以及不同浓度尼克酸对黑素瘤细胞的影响。应用RT-PCR技术检测转录因子snail2、twist及粘附因子E-cardherin的表达。结果通过Transwell体外迁移实验我们发现,尼克酸在体外对B16-F10黑色素瘤细胞的迁移有明显的抑制作用,且呈浓度依耐关系,尼克酸可促进肿瘤细胞迁移的相关基因snail2和twist表达明显减少(P <0.01),而抑制肿瘤细胞迁移相关基因E-cardherin明显增加。结论尼克酸在体外通能抑制黑素瘤细胞的迁移,对其治疗提供一定的理论基础。 Objective To explore the prospect and mechanism of niacin in the treatment of malignant melanoma. Methods Through transwell migration experiment in vitro,the migration inhibition effect of niacin on B16-F10 melanoma cells was observed,as well as the effect of different concentrations of niacin on melanoma cells. The expression of transcription factor snail2, twist and adhesive factor E-cardherin was detected by RT-PCR. Results Through transwell in vitro migration experiments, we found that niacin significantly inhibits the migration of B16-F10 melanoma cells, and shows a concentration-dependent relationship. Niacin can significantly reduce the expression of snail2 and twist related genes in tumor cell migration(P < 0.01), while E-cardherin inhibiting tumor cell migration was significantly increased. Conclusion Niacin can inhibit the migration of melanoma cells in vitro and provide theoretical basis for its treatment.
