摘要
目的建立应用AFS-8220型双通道原子荧光法(AFS)快速测定血、尿、脑脊液中砷的方法。方法选择最佳仪器工作条件,采用硝酸-过氧化氢体系微波消解样本,碱溶液中和消解液后AFS快速测定血、尿、脑脊液中砷。结果砷浓度在0.0~15.0μg/L范围内线性良好,R=0.9993,血、尿、脑脊液样品加标平均回收率分别为96.5%~99.5%、95.8%~99.1%和98.0%~101.6%,精密度高(RSD<2.05%),最低检出限0.052μg/L。结论所建立方法预处理样品时间短,操作快速简单,试剂用量少,灵敏度及准确度高,用于砷接触者及砷剂治疗患者的砷浓度快速检测,也可满足日常门诊砷浓度检测需求。
Objective To establish a fast method for detecting arsenic in blood, urine and cerebrospinal fluid by AFS-8220 two-channel atomic fluorescence spectrometry(AFS). Methods The optimum working conditions of instruments were choosed, samples were treated by microwave in nitric acid-hydrogen peroxide system and rapid determination of arsenic in blood, urine and cerebrospinal fluid by AFS after alkali solution neutralize digestion solution. Results The calibration curve for arsenic showed a good form of linear relation when arsenic density were in 0.0~15.0 μg/L, R=0.9993, and the average recoveries of blood, urine and cerebrospinal fluid samples were 96.5% ~99.5%, 95.8% ~99.1% and 98.0% ~101.6%,respectively. The precision was high(RSD<2.5%). The detection limit was 0.052 μg/L. Conclusion The method has the advantages of short sample pretreatment time, fast and simple operation, less reagent consumption, high sensitivity and accuracy. It can be used for rapid monitoring of arsenic concentration in arsenic contacts and patients treated with arsenic reagents, and can also meet the needs of daily outpatient arsenic concentration detection.
