去甲肾上腺素对人肺癌细胞株H460增殖、迁移和侵袭能力的影响

摘要 目的:研究去甲肾上腺素(norepinephrine,NE)对人肺癌细胞株H460增殖、迁移和侵袭能力的影响。方法:体外培养人肺癌细胞株H460,用NE干预后,CCK-8法检测细胞增殖,划痕愈合实验评价细胞迁移能力,侵袭小室实验观察细胞侵袭力,Western Blotting法检测细胞p38MAPK磷酸化蛋白(Phos-p38)的表达。结果:10μmol/L NE可显著促进H460细胞增殖,增强细胞的迁移和侵袭能力,增加p38MAPK磷酸化蛋白水平,与空白对照组比较,差异均有极显著统计学意义(P<0.01),且均能被β-肾上腺能受体(β-AR)的阻断剂普莱洛尔(PRO)和p38MAPK抑制剂U0126逆转。结论:NE能促进H460细胞的增殖、迁移和侵袭能力,其机制与p38MAPK通路激活有关。 AIM:To investigate the effect of norepinephrine(NE)on proliferation,migration and invasion of human lung cancer cell line H460.METHODS:Human lung cancer cell line H460 were cultured in vitro and treated with NE,then CCK-8mehod,scratch wound healing assay and transwell assay were applied to measure cell proliferation,migration and invasion.The expression of p38 MAPK phosphorylation(Phos-p38)were examined by Western blotting.RESULTS:Compared with the control,10μmol/L NE significantly enhanced proliferation,migration and invasion of human lung cancer cellline H460,and elevated p38 MAPK phosphorylation level(P<0.01).These effects were completely reversed byβ-adrenergic receptor(β-AR)antagonist propranolol(PRO)or p38 MAPK inhibitor U0126.CONCLUSION:NE could stimulate lung cancer cell proliferation,migration and invasion,its mechanism may be related with activation of P38 MAPK pathway.
