摘要 利用ISSR分子标记技术,对来自不同地区的25种无花果种质资源进行聚类分析,研究其遗传多样性并进行种质资源鉴定。以25种无花果叶片为材料,CTAB法提取其基因组DNA,进行ISSR分析,利用软件DPS(V7.5)计算遗传距离,利用UPGMA方法进行聚类分析,构建25种无花果样品的系统进化树。结果表明:从27条引物中筛选出19条有效引物进行ISSR分析,扩增得到86条带,26条是单态的,60条是多态的,多态性程度为69.77%。25种无花果间的遗传距离在0.038-0.833之间,平均为0.605。通过构建群居亲缘关系的分子系统树表明25种无花果品种在遗传距离为0.73处被分成2类:布兰瑞克单独聚为一类,其他品种聚为一类。25种无花果品种间的遗传差异较小,相关性较高;ISSR可作为一种特异性的分子标记用于无花果的品种鉴定。 The genetic diversity and the identification of germplasm resources of 25 Ficus carica L.from different regions was analyzed using ISSR technology.The genome DNA was extracted from 25 figs,with ISSR-PCR,the genetic distance was calculated by DPS software and cluster analysis was conducted by UPGMA.The results showed that 20 primers were screened from 27 primers for ISSR analysis.19 effective primers amplified 86 bands in the ISSR amplification of which 26 bands were monomorphic,60 were polymorphic,and the polymorphism was 69.77%.The genetic distance was 0.038 to 0.833,the average was 0.605.25 figs were divided into 2 classes at the genetic distance of 0.73:Branki was one class alone,and the other species were grouped into another class.The genetic differences between these 25 fig varieties were small and the correlation was higher.ISSR can also be used as a specific molecular marker for fig identification.
