三种工艺提取丹参有效成分及丹酚酸B分离纯化研究 被引量:4

摘要 目的:建立同时提取丹参中丹参酮IIA与丹酚酸B两种主要活性成分的方法及丹酚酸B的纯化工艺。方法:以丹参酮IIA与丹酚酸B的提取率和药效实验中血液流变参数为评价指标,考察最佳提取方法;并进一步用大孔树脂优选出丹酚酸B的最优分离纯化工艺。结果:3种提取方法中,SFE-CO2萃取法对丹参酮IIA的提取率明显高于乙醇超声提取;3种提取液都具有较好的改善血液流变学的作用,组间比较,乙醇超声提取和SFE-CO2萃取无显著性差异,但均明显优于不同浓度乙醇提取。丹参中丹酚酸B的分离、纯化选用SIPI905大孔树脂,丹酚酸B的最大上样量(即比吸附量)为9.20mg/g,洗脱剂为20%乙醇,用量为150mL,总固形物得率为5.56%,丹酚酸B得量占固体物量的64.98%。结论:综合3种提取方法的提取率以及药效实验结果,考虑降低成本、简化工艺和质量稳定,宜采用乙醇超声提取;SIPI905大孔树脂具备的优良特性适用于丹参中丹酚酸B的分离、纯化。 Objective:To establish a method for simultaneous extraction of tanshinone IIA and salvianolic acid B from Salvia miltiorrhiza and the purification process of salvianolic acid B.Methods:Taking the extraction rate of tanshinone IIA and salvianolic acid B and the hemorheological parameters in the pharmacodynamic experiment as the evaluation indexes,the optimum extraction method was investigated,and the optimum separation and purification process of salvianolic acid B was optimized by macroporous resin.Results:The extraction rate of SFE-CO2 was significantly higher than that of ethanol ultrasonic extraction in extracting tanshinone IIA,and the three extracts had better effect on improving hemorheology.There was no significant difference between groups in ethanol ultrasonic extraction and SFE-CO2 extraction,but they were obviously superior to ethanol extraction of different concentrations.Salvianolic acid B in Salvia miltiorrhiza was separated and purified by SIPI905 macroporous resin.The maximum sample loading(i.e.specific adsorption capacity)of salvianolic acid B was 9.20 mg/g,eluent was 20%ethanol and the dosage was 150 mL.The total solid yield was 5.56%,and salvianolic acid B accounted for 64.98%of the solid content.Conclusion:Ethanol ultrasonic extraction is suitable for the three extraction methods,considering the cost reduction,simplified process and stable quality,by synthesizing the extraction rate and pharmacodynamic test results of the three extraction methods.SIPI905 macroporous resin has excellent properties and is suitable for the separation and purification of salvianolic acid B from Salvia miltiorrhiza.
