摘要
将脂质体作为模拟生物膜,采用平衡透析法与紫外分光光度法研究附子提取液中乌头碱与脂质体的相互作用,并考察了薄膜分散法、乙醇注入法、缓冲液的pH值以及胆固醇和磷脂的比例对乌头碱与脂质体相互作用的影响。结果显示不同制备方法、脂质体的浓度、环境的pH值都对乌头碱与脂质体的相互作用有一定的影响。脂质体平衡透析法可用于预测有效成分在体内的吸收情况,为其药效物质基础的进一步研究提供依据。
Using balanced dialysis followed by UV analysis the water-soluble constituents in aconite which interacted with liposomes( as a simulate biomembrane) were analyzed. The film dispersion method and ethanol injection method were applied in this experiment. The effect of different pH values and the ratios of cholesterol to phospholipids were also investigated on their interactions. The interaction of aconitine with the liposome biomembrane was different under different conditions. The concentration of liposomes,preparation methods and the pH value of buffer affected the absorption of aconitine. Liposome equilibrium dialysis system could estimate the absorption of aconitine in vivo,and will provide evidence for the further study of the effective substance in aconitine.
