摘要
脂联素是由脂肪细胞特异性分泌的内源性细胞因子,通过与其受体Adipo R1、Adipo R2和T-cadherin结合,可激活其下游AMPK与PPAR-α等信号通路,在机体中发挥调节能量代谢、改善胰岛素抵抗、抗炎、抗氧化等多种生理功能。脂联素及其受体与胰岛素抵抗、心血管疾病、癌症、早老性痴呆、肝脏疾病等多种疾病的发生发展关系密切,是疾病临床治疗的新靶点。因此,脂联素及其受体在相关疾病治疗药物的研究中具有重要的参考价值。该文综述了脂联素及其受体的结构特征、表达调节、在相关疾病中作用及临床治疗策略的研究进展情况,以期为相关疾病的治疗与药物开发提供新思路。
Adiponectin is a endogenous cytokine secreted by adipocytes. Adiponectin plays important physiological roles in the human organism,such as regulation of energy metabolism,ameliorating insulin resistance,anti-inflammatory,antioxidant and so on with binding to its receptors via activation of AMPK and PPAR-α pathways. Adiponectin and adiponectin receptors was closely related to the occurrence and development of insulin resistance,cardiovascular disease,cancer,alzheimer' s disease and liver diseases. Adiponectin and adiponectin receptors might be a novel therapeutic target in clinical treatment of diseases,and have important reference values in pharmacy research for related diseases clinical treatment. The research progresses in the structure,expression regulation,effects in related disease and clinical treatment strategy of adiponectin and adiponectin receptors have been reviewed in this article.
