微小RNA-181b在脓毒症急性肺损伤大鼠中的表达及炎性机制研究 被引量:2

Expression of microRNA-181b in Sepsis-Induced Acute Lung Injury Rats and Its Correlation with the Levels of Inflammatory Cytokines

摘要 探讨微小RNA-181b(miR-181b)在脓毒症诱导急性肺损伤(Acute lung injury,ALI)发生时的差异表达及其与炎性因子白细胞介素-17(IL-17)表达的相关性。将24只28日龄健康雄性SD大鼠随机等量分为正常对照组、脓毒症3,6,12 h时相组(各6只),腹腔注射脂多糖(LPS) 10 mg/kg构建脓毒症大鼠ALI模型,采用HE染色观察大鼠肺组织病理形态变化。应用逆转录-聚合酶链反应(RT-PCR)研究SD大鼠ALI肺组织和对照组miR-181b的表达情况,采用酶联免疫吸附试验(ELISA)测定大鼠肺组织IL-17的含量并进行相关性分析。结果表明:HE染色示脓毒症组大鼠肺组织呈ALI改变;脓毒症各组大鼠肺组织miR-181b表达水平均明显低于正常对照组,差异均有统计学意义(P <0. 05);各实验组大鼠肺组织中miR-181b的表达与IL-17水平均呈高度负相关(r=-0. 878,P=0. 000)。说明miR-181b在脓毒症大鼠诱导ALI发生时的表达量下调,并与炎性因子IL-17表达水平呈负相关,有望成为脓毒症ALI早期诊断、治疗、预后评估的新生物学标志。 To investigate the differential expression of microRNA-181 b in sepsis-induced ALI and its correlation with IL-17,twentyfour SD male rats were randomly divided into 4 groups:normal control group,3-hour sepsis group,6-hour sepsis group,and 12-hour sepsis group in equal number.Sepsis-induced ALI model was induced by intraperitoneal injection of lipopolysaccharide(LPS,10 mg/kg).The injury of lung tissue was transcription polymerase chain reaction(RT-PCR),and IL-17 was measured by enzyme linked immunosorbent assay(ELISA)and the correlation analysis was carried out.The lung tissue hematoxylin-eosin staining indicated ALI in the sepsis group.The expression of miR-181 b in the lung tissue of each group of sepsis was significantly lower than that of the normal control group(P<0.05).The expression of miR-181 b in the lung tissue of the sepsis groups was negatively correlated with the level of IL-17(r=-0.878,P=0.000)in sepsis-induced ALI,and had a significantly negative correlation with IL-17,which may be used as a new biomarker of diagnostic,prognosis evaluation and therapy of sepsis-induced ALI.
