NucliSens EasyQ方法在HIV-1病毒载量检测中的应用及质量控制

摘要 利用NucliSens EasyQ核酸扩增检测分析系统对193例新发现的HIV感染者的病毒载量进行检测,以加强NucliSens EasyQ方法在HIV感染者中的诊断分析,并提高检测的精确度。并分析了患者血浆中CD4+T淋巴细胞与病毒载量的相关性,对HIV感染者体内免疫系统情况做初步的分析,并对该方法的检测结果和质量控制进行了分析。HIV感染者血浆中的病毒载量变化范围大,新发现的HIV感染者血浆中病毒载量为1. 98~7. 12 log/m L,平均值为4. 86 log/m L,与患者所处的HIV不同感染阶段以及自身的免疫能力密切相关。CD4+T淋巴细胞计数表明CD4+T淋巴细胞含量与HIV病毒载量呈负相关,当CD4+T淋巴细胞含量升高时,HIV病毒载量呈现降低的趋势,线性回归方程为log(Y)=-0. 003X+4. 583,皮尔森相关系数为r=-0. 384,P <0. 01。进一步对该方法的质量控制进行了分析,结果表明Nucli Sens Easy Q方法在HIV-1病毒载量检测中不仅高度有效,而且检出率高、误差小。①HIV感染者血浆中的病毒载量变化范围大,CD4+T淋巴细胞含量与HIV病毒载量呈负相关,可以考虑在临床上对HIV病毒载量及CD4+T淋巴细胞含量的相关性进行分析,从而更加精确地把握HIV感染者的病情发展,为HIV感染者的治疗提供准确的诊断数据;②HIV病毒载量测定采取严格的质量控制措施可以提高检测精确度。 In this paper,the viral load of 193 newly discovered HIV-infected persons was detected by Nucli Sens Easy Q nucleic acid amplification detection and analysis system,so as to enhance the diagnostic analysis of the Nucli Sens Easy Q method in HIV-infected persons and improve the accuracy of detection.The viral load of 193 newly discovered HIV-infected persons was detected by nucleic acid amplification detection and analysis system,and the correlation between CD4+T lymphocyte and viral load in plasma was analyzed.The immune system of HIV-infected persons was preliminarily analyzed,and the detection results and quality control of the method were analyzed.The viral load in plasma of HIV-infected persons varied widely.The newly discovered viral load in plasma of HIV-infected persons ranged from 1.98 to 7.12 log/m L,with an average of 4.86 log/m L,which was closely related to the different stages of HIV infection and their immune capacity.CD4+T lymphocyte count showed that CD4+T lymphocyte content was negatively correlated with HIV viral load.When CD4+T lymphocyte content increased,HIV viral load showed a decreasing trend.The linear regression equation was log(Y)=-0.003 X+4.583,Pearson correlation coefficient was r=-0.384,P<0.01.The quality control of this method was further analyzed.The results showed that Nucli Sens Easy Q method was not only highly effective in HIV-1 viral load detection,but also had high detection rate and small error.The viral load in plasma of HIV-infected persons varied widely,and the CD4+T lymphocyte content was negatively correlated with the HIV viral load.We can consider the correlation between the HIV viral load and the CD4+T lymphocyte content in clinical analysis,so as to grasp the development of HIV-infected patients more accurately and provide accurate diagnostic data for the treatment of HIV-infected persons;and HIV viral load measurement should be adopted.Strict quality control measures can improve the detection accuracy.
