摘要
目的探究秋水仙素的浓度和处理时间对淋巴细胞染色体制备的影响。方法研究不同秋水仙素终浓度(0. 2μg/ml,0. 3μg/ml,0. 4μg/ml)在不同秋水仙素处理时间(6 h,8 h,10 h)作用下对染色体中期分裂相的影响。结果秋水仙素终浓度和秋水仙素处理时间的交互作用对分裂指数(F=11. 982,P=0. 000 <0. 05)和分裂相合格率(F=4. 632,P=0. 004 <0. 05)影响差异有统计学意义。通过简单效应分析和事后多重比较分析,0. 3μg/ml秋水仙素处理8 h后,染色体分裂指数和分裂相合格率均较高,分别为58. 8%和55. 8%。结论对秋水仙素终浓度和处理时间定量控制,试验重现性良好,染色体中期分裂相数目充盈,形态良好,染色体畸变分析更准确客观。
Objective To explore the effect of colchicine’ s final concentration and action time on chromosome preparation of lymphocyte. Methods The effects of different final concentrations of colchicine( 0. 2 μg/ml,0. 3 μg/ml,0. 4 μg/ml) on the metaphase division of chromosomes under different colchicine action time( 6 h,8 h,10 h) were studied. Results The interaction between colchicine final concentration and action time had a statistically significant difference on mitotic index( F = 11. 982,P = 0. 000 < 0. 05) and qualified rate of metaphase chromosome spreading( F = 4. 632,P = 0. 004 < 0. 05). By the means of simple effect analysis and post hoc test,the best experimental condition is that with adding 0. 3 μg/ml colchicine in lymphocyto culturation for 8 h,the mitotic index is 58. 8%,and the qualified rate of metaphae chromosome spreading is 55. 8%. Conclusion Quantitative control measures make responses with stability and repeatablity. Good form of metaphase chromosome spreading can provide a more accurate diagnosis for chromosome aberration.
