不同照射方式下核黄素-A波紫外线交联作用的安全性及对兔巩膜成纤维细胞MMP-2表达影响的研究 被引量:2

摘要 目的探讨两种不同照射功率和照射时间核黄素A-波紫外线交联术对巩膜细胞的生物学作用及对兔巩膜成纤维细胞基质金属蛋白酶(matrix metallopeptidase 2,MMP-2)表达的影响。方法将实验兔随机分为A、B两组,右眼作为对照眼,不作任何处理;左眼作为实验眼,麻醉后暴露鼻上方巩膜,A、B组采用不同的照射方式,术后1周进行全视野视网膜电流图(full-field electroretinogram,f-ERG)检测。术后60 d培养眼交联部位巩膜成纤维细胞,检测MMP-2蛋白的表达。结果 A、B两组实验兔中,与对照眼相比,实验眼的暗、明适应3. 0的振幅和潜伏期差异无统计学意义(P> 0. 05)。A、B两组交联眼巩膜成纤维细胞中MMP-2的表达量明显高于对照眼,差异有统计学意义(t=8. 57、6. 38,P <0. 05); A、B两组交联眼之间比较,A组巩膜成纤维细胞中MMP-2的表达量明显高于B组,差异有统计学意义(t=2. 99,P <0. 05)。结论总能量相同的两种不同照射功率和照射时间核黄素A-波紫外线交联,均可加强巩膜组织的生物抗张力作用,具有较好的安全性。 Objective To To explore the biological effects of riboflavin A-wave ultraviolet cross-linking on scleral cells with two different irradiation powers and irradiation time,and its effect on the expression of matrix metallopeptidase 2( MMP-2) in rabbit scleral fibroblasts. Methods The experimental rabbits were randomized into group A( 12) and B( 12). For each rabbit,the right eye was chosen as the control group,while the left eye was the experimental group. After anesthesia,the nasal superior part of the sclera fibroblasts was fully exposed. Two different irradiation methods were used in A/B group. Both eyes were evaluated with the full-field electroretinogram( ERG) after CXL 1 week. 60 days after CXL,scleral fibroblasts were cultured from the CXL-treated sclera area and control sclera to investigate the expressions of MMP-2 protein. Results The ERG results showed that the differences of amplitude and latency in dark/light-adapted 3. 0 in all groups were not statisticallty significant( P > 0. 05). Compared with control eyes,the expression of MMP-2 in CXL fibroblasts of A and B group increased significantly,which were both have statistical significance( t = 8. 57,6. 38,P < 0. 05). Compared with B group,the MMP-2 expression level of A CXL group is significantly higher. The difference was statistically significant.( t = 2. 99,P < 0. 05). Conclusion The two different irradiation powers with the same total energy and the irradiation time of riboflavin A-wave ultraviolet cross-linking can enhance the bio-tension resistance of scleral tissue and have better safety.
