摘要
目的通过对青海省脊髓灰质炎(脊灰)实验室2016-2017年两年的细胞敏感性(cell sensitivity,CS)监测结果进行分析,评估其在脊灰实验室检测质量控制中的重要作用。方法统一使用96孔微量板病毒滴定法在表达脊灰病毒受体的小鼠肺(Mouse Lung Cells that Express Receptors for Human Polioviruses,L20B)细胞和人横纹肌肉瘤(Rhabdomyosarcoma,RD)细胞上对Ⅰ型及Ⅲ型,两个型别的脊灰病毒的敏感性进行检测。结果 2016-2017年共开展8次常规CS监测并按时向国家脊灰实验室上报结果,CS监测结果显示青海省脊灰实验室的实验室质量控制(Quality control of laboratory,LQC)株滴度均在正常范围±0. 5logCCID50/0. 1 ml波动。结论青海省2016-2017年脊灰实验室所用的L20B和RD细胞对脊灰病毒敏感性处于正常范围内。CS监测作为中国脊灰实验室网络(Chinese Poliomyelitis Laboratory Network,CPLN)质量控制的重要指标,保证了实验室检测脊灰病毒的敏感性。
Objective To analyze the cell sensitivity(CS)monitoring results of poliomyelitis laboratory in Qinghai Province from 2016 to 2017,and evaluate the important role in the quality control of poliomyelitis laboratory testing.Methods The sensitivity for two types of poliovirus in Mouse Lung Cells that Express Receptors for Human Polioviruses and Rhabdomyosarcoma was tested by using 96 microplate virus titration.Results A total of eight conventional CS monitoring were performed from 2016 to 2017 and the results were reported to National Poliomyelitis Laboratory on time,and CS monitoring results showed that the titer of quality control of laboratory(LQC)of polio strains in Qinghai Province were fluctuated within the normal range of 0.5 logCCID50/0.1 ml.Conclusion L20 B and RD cells used in the polio laboratory of Qinghai Province were within the normal range of sensitivity to poliovirus in 2016-2017.As an important indicator of quality control of the Chinese Poliomyelitis Laboratory Network(CPLN),CS monitoring ensures the sensitivity of Laboratory detection of poliovirus.
