摘要
目的对采自石家庄地区的小家鼠肠道内的寄生虫进行形态描述,依据形态学特征和分子生物学信息进行鉴定。方法按照不同的寄生部位(皮下、肌肉、消化道、肝脏、心脏、体腔)对鼠进行解剖,将检得的蠕虫分别放入不同的培养皿中,然后用80℃左右的热水将虫体烫直,随后保存在70%的酒精中。在光学显微镜和扫描电镜下观察、测量、拍照,并对其ITS-1、ITS-2、18S rRNA、28S rRNA基因扩增并测序。结果依据所采线虫的形态学特征鉴定其为四翼无刺线虫,测序结果进一步证实所采线虫为四翼无刺线虫。结论本文通过光学显微镜和扫描电镜(SEM)的观察,详细描述了四翼无刺线虫的形态特征,对其4个基因测序,测序结果经比对进一步证实为四翼无刺线虫。
Objective Morphological descriptions of parasites in the intestines of Mus musculus from Shijiazhuang were carried out and identified based on morphological characteristics and molecular biological information.Methods The rats were dissected according to different parasitic parts(subcutaneous,muscle,digestive tract,liver,heart,body cavity),and the detected worms were placed in different culture dishes,and then the worms were hotted with hot water of about 80℃to straight,then stored in 70%alcohol.The specimens were observed and measured by light microscopy and scanning electron microscopy(SEM).In addition,the ITS-1,ITS-2,18S rRNA,28 S rRNA genes were amplificated and sequenced.Results All specimens collected from Mus musculus belong to Aspiculuris tetraptera based on morphological characteristics and molecular biological information.Conclusion The worms were observed by light microscopy and scanning electron microscopy(SEM).Detailed characteristics of Aspiculuris tetraptera were described.In addition,4 genes were sequenced and sequencing results also confirmed our specimens belong to Aspiculuris tetraptera.
