褐家鼠寄生鼠异刺线虫形态描述与分子鉴定

Morphological description and molecular identification of Heterakis spumosa from Rattus norvegicus

目的对采自石家庄地区的褐家鼠消化道内的线虫进行形态学描述,根据形态学特征和基因信息进行鉴定。方法对褐家鼠进行解剖,根据不同的采集部位将检得的寄生虫放入不同的容器中,使用75~85℃的水将虫体烫直,放入70%的酒精中保存。使用光学显微镜观察并测量,对特殊结构拍照,并对其COXI、COXII、ITS-2、18S rRNA、28S rRNA基因扩增并测序。结果依据所采线虫的形态学特征初步鉴定为鼠异刺线虫,测序结果进一步证实所采线虫为鼠异刺线虫。结论本文通过光学显微镜的观察,详细描述并绘制了鼠异刺线虫的形态特征,对其5个基因测序,测序结果经比对进一步证实为鼠异刺线虫。

Objective Morphological description of nematodes in the digestive tract of Rattus norvegicus from Shijiazhuang is to be made.To identify based on morphological characteristics and genetic information.Methods The Rattus norvegicus were dissected,and the detected nematodes were placed in different containers according to different collection sites.The nematodes were scalded with water at 75~85°C and stored in 70%alcohol.The specific structure was photographed and measured using an optical microscope,and the COXI,COXII,ITS-2,18 S rRNA,and 28 S rRNA genes were amplified and sequenced.Results According to the morphological characteristics of the nematodes,the elegans were identified as Heterakis spumosa,which was further confirmed by the sequencing results.Conclusion In this paper,the morphological characteristics of Heterakis spumosa in the digestive tract of Rattus norvegicus were described and mapped by light microscopy.There are 5 genes were sequenced and were further confirmed by the sequencing results.
