摘要
目的:研究可溶性CD40配体(Soluble CD40Ligand,sCD40L)对人白血病THP-1细胞增殖的作用,并探讨其对生存素mRNA(survivin mRNA)及白细胞介素6(IL-6)表达的影响。方法:采用MTT法检测sCD40L孵育THP-1细胞体外增殖的影响;通过RT-PCR检测survivin mRNA表达;ELISA测定IL-6的影响。结果:不同浓度的sCD40L对THP-1细胞增殖有抑制作用,并呈剂量-时间依赖关系;sCD40L对THP-1细胞survivin及IL-6的表达有明显的抑制作用,并呈剂量-时间依赖性。结论:sCD40L在体外能够明显抑制THP-1细胞增殖,具有剂量-时间依赖性。sCD40L抑制THP-1细胞增殖与survivin及IL-6表达相关,并可能与其下调有关。
Objective:To investigate the effect of sCD40 Lon proliferation of THP-1cells and expressions of IL-6and survivin.Methods:The MTT assay was used to examine the effect of different concentrations of sCD40 Lon Leukemia THP-1cells proliferation in vitro at different incubation times;RT-PCR was used to determine the effect of different concentrations of sCD40 Lon the expression of survivin mRNA in THP-1cells at different incubation times;ELISA was used to detect changes in cytokine IL-6levels in the culture supernatants of leukemia THP-1cells of different concentrations of sCD40 Lat different incubation times.Results:Different concentrations of sCD40 Lcould all significantly inhibit THP-1cells proliferation and there was a dose-dependent tendency.sCD40 Lcould suppress the expression of survivin and IL-6,and with dose-dependent manner.Conclusion:sCD40Lcan significantly inhibit the proliferation of THP-1cells in vitro,yet it is concentration and time dependent;the mechanisms of sCD40 Lin inhibiting the proliferation of THP-1cells and inducing apoptosis in THP-1cells may be related to their inhibition of IL-6and survivin expression.
