^(60)Coγ射线全身照射后大鼠血浆中GDF-15、Flt-3L和SAA表达的剂量相关性研究

摘要 目的:探讨钴60(^(60)Co)γ射线照射对SD大鼠血浆中Fms样酪氨酸激酶3配体(Flt-3L)、血清淀粉样蛋白A(SAA)和生长分化因子-15(GDF-15)蛋白表达情况的影响。方法:将72只SD大鼠按照不同照射剂量分为0 Gy组、1 Gy组、3 Gy组和5 Gy组,每组18只,采用^(60)Coγ射线照射大鼠,剂量率为1 Gy/min。照射后1 d、3 d和5 d采集4组大鼠外周血,通过酶链免疫吸附试验(ELISA)检测血浆中的Flt-3L、SAA和GDF-15蛋白表达水平。结果:SD大鼠血浆中的Flt-3L和GDF-15蛋白在照射后表达升高,升高的水平与辐射剂量相关。3 Gy组和5 Gy组血浆中的GDF-15蛋白在照射后1 d、3 d和5 d与0 Gy组相比表达均增高,差异有统计学意义(t=3.09,t=6.32,t=8.91,t=10.71,t=2.73,t=6.64;P<0.05);5 Gy组血浆中的Flt-3L蛋白在照后1 d、3 d和5 d与0Gy组相比表达均增高,差异有统计学意义(t=4.22,t=8.48,t=5.51;P<0.05);3 Gy组Flt-3L蛋白只在照后1 d和5 d表达增高,差异有统计学意义(t=5.79,t=3.69;P<0.05);GDF-15蛋白水平在照射后1 d的剂量-效应关系(R^2=0.936,P<0.05)优于照射后3 d和5 d(R^2=0.913,R^2=0.887;P<0.05);Flt-3L蛋白正相反,其在照射后5 d的剂量-效应关系最好(R^2=0.965,P<0.05);SAA只有在照射后1 d有随剂量升高表达增加的趋势。结论:血浆中的Flt-3L和GDF-15蛋白在全身照射的大鼠血浆中特定时间点呈现出比较良好的剂量-效应关系。 Objective:To investigate the influence of Coγ-rays irradiation on the expression of plasma Flt-3L,SAAand GDF-15 of SD rats.Methods:A total of 72 SD rats were divided into 0 Gy group,1 Gy group,3 Gy group and 5Gy group in accordance with different irradiation dose,and 18 rats in each group.And rats were irradiated with 60Coγ-rays at a dose rate of 1 Gy/min.And peripheral bloods of all of rats were collected at 1d,3d and 5d post irradiation,and enzyme linked immune sorbent assay(ELISA)was used to detect the expression levels of plasma Flt-3L,SAA and GDF-15 proteins.Results:The expressions of plasma Flt-3L and GDF-15 protein increased after irradiation,and the increased level was relative with radiation dose.Compared with that of 0Gy group,the expressions of plasma GDF-15of 3 Gy group and 5 Gy group at 1d,3d and 5d post irradiation were significantly increased(t=3.09,t=6.32,t=8.91,t=10.71,t=2.73,t=6.64,P<0.05).Compared with that of 0Gy group,the expressions of plasma Flt-3L of 5 Gy group at 1d,3d and 5d post irradiation were significantly increased(t=4.22,t=8.48,t=5.51,P<0.05).And the expressions of plasma Flt-3L of 3 Gy group only significantly increased at 1 d and 5 d post irradiation(t=5.79,t=3.69,P<0.05).The dose-response relationship of GDF-15 protein at 1d post irradiation(R^2=0.936,P<0.05)was better than that at 3d post irradiation and at 5d post irradiation(R^2=0.913,R2=0.887,P<0.05),respectively.However,the best dose-response relationship of Flt-3L protein occurred at 5 d post irradiation(R^2=0.965,P<0.05).Besides,the tendency that SAAexpression increased with the enhancing of dose only occurred at 1d post irradiation.Conclusion:The expressions of plasma Flt-3L and GDF-15 in rats that receive whole body irradiation have a relatively good dose-response relationship at specific time points.
