核苷酸和核苷酸类药物对癌症基因组稳定性作用比较 被引量:1

摘要 目的总结国内外对核苷酸及有关核苷酸类药物对肿瘤基因组不稳定性作用的研究现状。方法检索Medline和全国图书馆参考咨询联盟、万方医学网(中文和英文数据库均需检索),1998年1月—2015年5月发表的、关于核苷酸及核苷酸类药物对肿瘤基因组不稳定性作用的对照研究。遵循严格的纳入标准和剔除标准,提取文献的数据进行分析。结果共检索到英文文献7 165篇,中文文献53篇。最后纳入分析的英文文献36篇,中文文献2篇。在检测到的28篇英文文献中,5篇主要是表明多数恶性肿瘤细胞存在染色体及其基因组的不稳定性,并解释其原因与癌细胞的DNA复制时的应激状态相关。2篇文献的研究涉及低核苷酸水平对基因组的不稳定性。2篇文献的研究涉及补充外源性核苷酸对基因组具有稳定性的作用。6篇文献的研究证明,激活核苷酸的生物合成通路,补救了复制应激和基因组的不稳定性。7篇,是关于核苷酸类药物及最近开发的新药的抗癌机制,一般是通过并入癌细胞中的DNA链而阻断DNA合成,并可能导致癌细胞凋亡。3篇英文文献主要涉及癌细胞的抗药作用。2篇中文文献表明,8-氯腺苷阻滞肿瘤细胞在G2/M期,并由此激活凋亡信号通路而诱导细胞凋亡。结论核苷酸类药物嵌入DNA链后导致基因组变化呈多样性,对于癌症有治疗价值,也有不可忽视的并发症。
