百瑞口服液对阿霉素肾病大鼠综合征的治疗作用

Therapeutic Effects of Bairui Oral Liquid on Adriamycin-Induced Nephropathy Rats

目的观察百瑞口服液对阿霉素(通用名注射用盐酸多柔比星,ADR)诱导的肾病综合征大鼠的治疗作用。方法将60只SD大鼠,随机均分为空白对照组,模型对照组,阳性对照组(醋酸泼尼松5.0 mg/kg),百瑞口服液低剂量组(0.5 mL/kg,相当于每1 kg体重0.25g生药)、中剂量组(2 mL/kg,相当于每1 kg体重1.0 g生药)、高剂量组(10 mL/kg,相当于每1 g体重5.0 g生药)。除空白对照组外,一次性尾静脉注射阿霉素6.0 mg/kg,次日连续灌胃给药6周。观察药物对肾病大鼠尿量、24 h尿蛋白、血液生化指标及肾组织病理学等的影响。结果与模型对照组相比,各给药组在治疗第4周,尿量均开始增多,百瑞口服液中、高剂量组尿量显著升高(P<0.01)。与模型对照组相比,给药第5周后,百瑞口服液高剂量组尿蛋白下降(P<0.05),阳性对照组和百瑞口服液中剂量组尿蛋白显著下降(P<0.01),低剂量组则无差异。与模型对照组相比,阳性对照组的血清总蛋白(TP)水平升高,总胆固醇(TCH)和三酰甘油(TG)水平降低(P<0.05),百瑞口服液中剂量组的TP,血清白蛋白(ALB)水平升高(P<0.05),高剂量组TCH和TG水平降低(P<0.05)。结论百瑞口服液能够减轻SD大鼠肾病综合征烂尾、水肿等症状,具有利尿、降低24 h尿蛋白、提高血清蛋白水平、调血脂等作用,能改善病理组织学变化,减轻肾脏损害,对肾病综合征大鼠具有良好的治疗效果。

Objective To observe the therapeutic effects of Bairui Oral Liquid on adriamycin-induced nephropathy rats.Methods Sixty SD rats were randomly and equally divided into the blank control group,model control group,positive control group(prednisone acetate 5.0 mg / kg),low-dose group(0.5 mL / kg,equivalent to 0.25 g crude drug per 1 kg body weight),middle-dose group(2 mL / kg,equivalent to 1.0 g crude drug per 1 kg body weigh) and high-dose group(10 mL / kg,equivalent to 5.0 g crude drug per 1 kg body weight).Except the blank control group,the other groups were given once adriamycin 6.0 mg / kg by the tail intravenous injection.From the next day,the gavage medication was given for successively 6 weeks.The influences of medication on the urine volume,24 h urine proteins,blood biochemical indexes and the renal histopathology were observed.Results Compared with the model group,the urine volume in 4 weeks after therapy in various medication groups began to increase,which in the middle-dose and high dose groups was significantly increased(P < 0.01).Compared with the model group,the urine protein after 5 weeks of medication in the high-dose group was decreased(P < 0.05),which in the positive control and the middle-dose groups were significantly decreased(P < 0.01) and which in the low-dose group had no difference.Compared with the model group,serum total protein(TP) in the positive control group was increased,while the total cholesterol(TCH) and triglyceride(TG) levels were decreased(P < 0.05),TP and albumin(ALB) in the middle-dose group were increased(P < 0.05),the TCH and TG levels in the high-dose group were decreased(P < 0.05).Conclusion Bairui Oral Liquid can alleviate the symptoms of tail necrosis and tissue edema,etc.,and has the effects for diuresis,reducing 24 h urine protein,increasing the albumin level and regulating blood lipid,which can improve the histopathological changes,relieve the renal damage and has the better curative effects on the rats with nephrotic syndrome.
