高效液相色谱法测定七叶一枝花酊中盐酸小檗碱含量

Determination of Berberine Hydrochloride in Paris Polyphylla Tincture by HPLC

目的建立测定七叶一枝花酊中盐酸小檗碱含量的高效液相色谱(HPLC)法。方法色谱柱为Wondasil C18柱(150 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(含4 g/L庚烷磺酸钠,40∶60),流速为1.0 m L/min,柱温为30℃,检测波长为345 nm,进样量为10μL。结果盐酸小檗碱质量浓度在12.19~243.84μg/m L范围内与峰面积线性关系良好(r=0.999 9),平均加样回收率为100.84%,RSD为0.93%。结论该法简便、准确、重复性好,可作为七叶一枝花酊中盐酸小檗碱含量的测定方法。

Objective To establish an HPLC method for the determination of berberine hydrochloride in Paris Polyphylla Tincture. Methods The analytical column was Wondasil C18 column ( 150 mm × 4. 6 mm, 5 μm ) with acetonitrile-0. 1% phosphoric acid ( containing 4 g/L sodium heptanesulfonate,40:60 ) as mobile phase;the flow rate was 1. 0 mL/min, and the detecting wavelength was at 345 nm;The sample was loaded for 10 μL, and column temperature was set at 30℃. Results A good linearity of berberine hydrochloride was obtained within 12. 19-243. 84 μg/mL( r=0. 999 9);the average recoveries of berberine hydrochloride was 100. 84% and the RSD was 0. 93%. Conclusion The method is simple, accurate and repeatable, which can be used for quality control of berberine hydrochloride in Paris Polyphylla Tincture.
