紫杉醇参与逆转肺部肿瘤细胞免疫逃逸的机制研究 被引量:2

摘要 目的分析紫杉醇类抗肿瘤药物紫杉醇参与逆转肺部肿瘤细胞免疫逃逸机制,为治疗肺部肿瘤提供参考。方法采用Lewis肺癌细胞株和紫杉醇(5×108 mol/L)处理细胞株抗原肽转运体(TAP),通过流式单抗TAP1及TAP2进行体外试验检测,体内试验分为肿瘤组和化疗组,并设置空白对照组,化疗组小鼠采用Lewis肺癌细胞株经尾静脉注射成瘤,取小鼠肺部组织,采用流式三染法测定调节性T细胞(Treg)。结果经紫杉醇处理的Lewis肺癌细胞株紫杉醇组表面TAP1表达为(5.68±0.65)%,TAP2表达为(89.54±4.81)%,显著高于Lewis肺癌细胞株组的(1.93±0.25)%和(66.84±5.16)%(P<0.05);正常小鼠Treg细胞表达为(12.45±1.53)%,显著低于荷瘤小鼠肺部Treg细胞表达的(25.49±2.29)%(P<0.01);紫杉醇化疗组小鼠肺部Treg细胞表达为(17.52±2.28)%,显著低于荷瘤小鼠肺部Treg细胞表达(P<0.05)。结论紫杉醇类抗肿瘤药物紫杉醇可能通过消除肿瘤局部Treg的产生,部分参与逆转肺部肿瘤细胞免疫逃逸机制。 Objective To analyze immune escape mechanisms of paclitaxel involved in reversal lung cancer cells. Methods The cell line antigen peptide transporter( TAP) was processed adopted Lewis lung carcinoma cells lines and paclitaxel 5 × 108 mol / L,in vitro assay was performed by flow through monoclonal TAP1,TAP2. In vivo test was divided into the tumor group and the chemotherapy group,and control group was set. The chemotherapy group was adopted Lewis lung carcinoma cell lines via the tail vein injection into the tumor,the mice lung tissue was selected,the flow was determined by three- regulatory T cells staining( Treg). Results The TAP1 and TAP2 of Lewis lung cell surface treated by paclitaxel was( 5. 68 ± 0. 65) % and( 89. 54 ± 4. 81) %,which were significantly higher than( 1. 93 ± 0. 25) % and( 66. 84 ± 5. 16) % of the Lewis lung carcinoma cell line group,respectively( P < 0. 05),the Treg cells of normal mice was( 12. 45 ± 1. 53) %,which was significantly lower than( 25. 49 ± 2. 29) % of mice lung Treg cells( P < 0. 05); the Treg of cells paclitaxel mice lung was( 17. 52 ± 2. 28) %,which was significantly lower than that of mice lung cells( P < 0. 05). Conclusion Taxol anticancer drugs paclitaxel may eliminate tumors Treg by local regulatory T cells,and partly involved in reversing lung tumor cell immune escape mechanisms.
