摘要
目的建立测定参肤软膏中盐酸小檗碱含量的高效液相色谱(HPLC)法。方法色谱柱为Kromasil C18柱(250 mm×4.6 mm,5μm),流动相为0.1%磷酸(每100 m L加入十二烷基磺酸钠0.02 g)-乙腈(59∶41),流速为1.0 m L/min,检测波长320 nm,柱温室温,进样量20μL。结果盐酸小檗碱与其他组分分离良好,进样量在1.184~5.920μg范围内与峰面积线性关系良好(r=0.999 4,n=5);平均加样回收率为99.83%,RSD为0.30%(n=6)。结论该法简单、专属性强、重复性好,可用于参肤软膏中盐酸小檗碱的含量测定。
Objective To establish a content determination method of berberine hydrochloride in Shenfu Ointments. Methods HPLC analysis was carried out on a Kromsal C18 chromatographic column (250 mm × 4. 6 mm,5 μm),the mobile phase was acetonitrile-0. 1% phosphoric acid(59 :41)(adding 0. 02 g sodium dode-cylsulphate into 100 mL solution)with the flow rate of 1. 0 mL/min and the detection wavelength was 320 nm;the temperature of column was room temperature,and the injection volume was 20 μL. Results The linear range was 1. 184-5. 920 μg with good correlation with the peak area( r=0. 999 4,n=5). The average recovery was 99. 83% and RSD was 0. 30%( n=6). Conclusion The established method is accurate,specific and reproducible,and suitable for the determination of berberine hydrochloride in Shenfu ointments.
