三七总皂苷对脑缺血-再灌注损伤大鼠脑神经的保护作用及机制 被引量:4

Neuroprotective Role and Mechanism Research of Panax Notoginseng Saponins on Cerebral Ischemia Reperfusion Injury in Rats

摘要 目的探讨三七总皂苷对脑缺血-再灌注损伤大鼠脑神经的保护作用及作用机制。方法选取60只SD雄性大鼠,按随机数字表法将其分为假手术组(A组)、模型组(B组,线栓法建立大鼠大脑中动脉局灶缺血-再灌注损伤模型后,腹腔注射生理盐水)、三七总皂苷组(C组,造模后,腹腔注射三七总皂苷100 mg/kg),各20只。结果与A组相比,B组、C组神经症状评分、脑梗死体积均明显增高(P<0.05),依文思蓝、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)含量均明显提高(P<0.05);与B组相比,C组神经症状评分、脑梗死体积均明显降低(P<0.05),依文思蓝、IL-1β、TNF-α含量均明显减少(P<0.05)。结论三七总皂苷通过降低脑部细胞因子的表达、减少血脑屏障通透性,发挥对脑缺血-再灌注损伤大鼠的脑神经保护作用。 Objective To study the neuroprotective role and mechanism research of panax notoginseng saponins( PNS) on cerebral ischemia reperfusion injury in rats.Methods 60 male SD rats were randomly divided into the sham operation group( group A),model group( group B,ip with normal saline after ischemia reperfusion injury by ligation of middle cerebral artery) and the PNS group( group C,ip with 100 mg / kg PNS after model establishment),20 rats per group.Results Compared with the group A,the neurological deficits scores and brain infarct size in the group B and group C were significantly increased( P < 0.05); and the content of Evans blue was obviously improved in group B and group C( P < 0.05); compared with group B,the neurological deficits scores and brain infarct size were significantly decreased( P < 0.05) and the contents of Evans blue,IL- 1β,TNF- α were obviously reduced in group C( P < 0.05).Conclusion PNS can play the neuroprotective role on cerebral ischemia reperfusion injury in rats by decreasing the expression of cytokines in brain tissue and reducing the permeability of blood brain barrier.
