摘要
目的研究视黄酸(RA)促进脐血淋巴细胞(CBL)抗体生成的作用机制。方法对脐血标本进行血清维生素A(VA)浓度测定,采用 RT-PCR 法检测 CBL 中视黄酸受体-α(RAR-α)的基因表达水平,并采用 EL ISA 法检测细胞培养上清液中 IgM 含量。结果脐血血清 VA 水平与未培养 CBL 中 RAR-α的基因表达水平之间存在正相关 r=0.50(P<0.01);RA 增加金黄色葡萄球菌 Cow an I 株(SAC)活化 CBL 的 IgM 生成能力(P<0.001);RA 上调 SAC 活化 CBL 中 RAR-α的基因表达水平(P<0.001);RA 使 RAR-α基因表达的上调幅度与 RA 使 IgM 生成的上调幅度之间存在正相关:r=0.04(P<0.05)。结论 RA 对 CBL 抗体生成的促进作用可能通过 RAR-α途径传递。
Objective To further explore the mechanism by which retinoic acid(RA)promotes the antibody production of cord blood lymphocytes(CBL).Methods The levels of serum vitamin A in cord blood were measured. The expression of retinoic acid receptor-α(RAR-α)gene was determined by RTPCR(reverse transcription- polymerase chain reaction).The IgM concentrations in the supernatants of cell culture were measured by EL ISA. Results A positive correlation was found between serum vitamin A level and RAR-α expression level in uncultured CBL:r=0.50(P<0.01).IgM synthesis of SAC-activated CBL was augmented by RA(P<0.001).RAR-α expression level of SAC-activated CBL was increased by RA(P<0.001).RAR-α expression level of SAC-activated CBL was increased by RA(P<0.01).There was a positive correlation between the enhanced amplitude of RAR-α expression and the enhanced amplitude of IgM synthesis by RA:r=0.40(P<0.05).Conclusion Our studies suggest that RAR-α pathway perhaps is one of main ways by which RA promotes antibody production of CBL.
