摘要
目的应用蛋白芯片技术筛选并分析参苈加颗粒对慢性心力衰竭(以下简称心衰)大鼠心肌内分泌腺源性血管内皮生长因子(EG-VEGF)表达的影响。方法从70只清洁级Wistar大鼠中随机抽取15只作为空白组(仅给予生理盐水),其余55只用阿霉素制作心衰模型。6周后,将造模成功的45只大鼠随机分为治疗组(25只)和模型组(20只),分别给予参苈加颗粒和生理盐水灌胃,1次/d。4周后取大鼠左心室组织,用蛋白芯片技术分析实验大鼠标本EG-VEGF的表达。结果与空白组比较,模型组EG-VEGF的表达下调,相对丰度比为0.52;与模型组比较,治疗组EG-VEGF的表达上调,相对丰度比为0.66;治疗组与空白组相比表达无差异,相对丰度比为0.79。结论参苈加颗粒可通过调节EG-VEGF的表达,改善心肌纤维化程度,对心衰大鼠有一定的保护作用。
Objective To assess the effect of SHENLIJIA Granule on EG-VEGF expression in heart failure rats by the protein chips. Methods There were 70 Wistar rats in this experiment. Fifteen rats were randomly selected as blank group, the others were induced by intraperitoneal injection of doxorubicin. The survival rats were randomly divided into model and treatment group. After the model establishment. In the meanwhile, rats in the treatment group were treated with gastric perfusion of Chinese herb SHENLIJIA Granule, once a day for twenty days. And those in the blank and model groups were treated with gastric perfusion saline. Four weeks later, gathering the left cardiac muscular tissue for the protein chips. Results Compared with the blank group, the EG-VEGF expression in the model group was down and their relative abundance was 0.52. Compared with the model group, the EG-VEGF expression in the treatment group was up and their relative abundance was 0.66. There was no difference between the treatment group and the blank group, their relative abundance was 0.79. Conclusions SHENLIJIA Granule improved the degrees of myocardium fibrosis and protected the heart by raising the expression of EG-VEGF.
