摘要
目的检测FBLN1在舌鳞状细胞癌(tongue squamous cell carcinoma,TSCC)中表达、启动子区甲基化水平以及甲基化对肿瘤进展过程的影响。方法应用蛋白印记方法检测78例TSCC组织及配对癌旁组织中的表达,应用甲基化特异性聚合酶链反应(Methylation-specic polymerase chain reaction,MSP)技术检测FBLN1基因启动子区甲基化水平。应用SPSS 17.0统计软件进行数据分析,组间比较采用χ2检验和t检验,P<0.05为差异有统计学意义。结果 FBLN1基因在癌组织与正常组织中的相对表达量分别为(0.746±0.326)和(2.353±0.949),差异有统计学意义(t=2.245,P<0.05);FBLN1基因的低表达与肿瘤临床分期及肿瘤的分化程度密切相关(P<0.05)。癌组织基因启动子区甲基化发生率为41.0%(32/78),正常组织甲基化发生率为7.7%(6/78),差异有统计学意义(χ2=19.26,P<0.01)。结论 FBLN1基因的低表达与TSCC患者肿瘤临床分期及肿瘤细胞分化程度密切相关,其低表达与启动子区甲基化有关。
Objective To investigate the expression and promoter-methylation level of Fibulin-1 gene(FBLN1) in tongue squamous cell carcinoma(TSCC). Methods Western blot was carried out to detect the protein expression of FBLN1 for 78 pairs of TSCC and normal control tissues. FBLN1 promoter methylation status was measured using methylation-specic polymerase chain reaction(MSP) in 78 pairs of tissues. Results The relative expression of FBLN1 in TSCC tissues(0.746±0.326) was significantly lower than that of normal tissues(2.353±0.309)(t=2.245,P<0.05). Downregulation of FBLN1 expression was closely correlated with the degree of differentiation and clinical stage(P<0.05). MSP results indicated that the FBLN1 gene promoter methylation rates(41.0%) was significantly higher than that in normal tissue(7.7%)(χ2=19.26, P<0.01). Conclusions The lower expression of TSCC patients is closely related with TSCC clinical stage and tumor differentiation, and promoter methylation may cause its lower expression.
