摘要
目的探讨针刺百会、腰奇穴对戊四氮(PTZ)致癫痫大鼠惊厥潜伏期及海马区NO、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)的影响。方法清洁级大鼠90只,随机选取10只为空白组,其余通过PTZ建立癫痫大鼠模型,在造模成功的大鼠中随机选取50只分成模型组、非经非穴组、百会组、腰奇组、百会腰奇组,每组10只。除空白组和模型组外均予针刺治疗,治疗结束后记录大鼠惊厥潜伏期,采用分光光度法检测各组海马区NO含量及NOS、SOD活力。结果与模型组比较,百会组、腰奇组、百会腰奇组惊厥潜伏期延长(P均<0.05)。模型组大脑海马区NO含量及NOS、SOD活力与空白组比较明显降低,差异有统计学意义(P<0.01);与模型组比较,非经非穴组各指标差异无统计学意义(P>0.05),百会组、腰奇组、百会腰奇组NO含量及NOS、SOD活力明显升高,差异有统计学意义(P<0.01);百会组、腰奇组、百会腰奇组组间比较差异无统计学意义(P>0.05)。结论针刺百会或腰奇穴可以延长癫痫模型大鼠惊厥潜伏期,增加PTZ致痫大鼠海马区NO含量,提高海马区NOS、SOD活力。
Objective To investigate the effect of acupuncture of Baihui Point and Yaoqi Point on the latent period of convulsion, nitric oxide(NO), NO synthase(NOS) and superoxide dismutase(SOD) in hippocampus of epileptic rats. Methods This study enrolled 90 rats of clean grade, in which 10 cases of rats were acted as blank group, the rest rats were used to establish epileptic rats model by PTZ.Fifty epileptic rats were randomly divided into five groups of 10, namely model group, the non-acupoint group, the Baihui group, the Yaoqi group, and the Baihui-Yaoqi group. The acupuncture were performed in these groups except the blank group and the model group. And then observing the latent period of the rats, using the spectrophotometric method to test the contents of NO and the vitality of NOS, SOD in the rat's hippocampus after treatment. Results Comparing to the blank group, the latent period of the rats was prolonged after the acupuncture in the Baihui group, the Yaoqi group, and Baihui group-Yaoqi group(all P<0.05). The content of NO and the vitality of NOS, SOD of the rat's hippocampus were lower in the model group than in the blank group(all P<0.01). There were higher NO level and vitality of NOS,SOD in the Baihui group, the Yaoqi group, and Baihui groupYaoqi group than in the model group, but there was no significant difference of above indexes between the model group and the non-acupoint group, the same resuls was obtatined among the Baihui group,the Yaoqi group, and Baihui group- Yaoqi group(all P>0.05). Conclusions Acupuncture of Baihui Point and Yaoqi Point can make the latent period of convulsion prolonged, the level of NO increased, the activity of NOS, SOD in hippocampus enhanced in the epileptic rats.
