摘要
目的探讨不同剂量维生素C(vitamin C,VC)联合低剂量维生素D_3(vitamin D_3,VD_3)对葡聚糖硫酸钠(dextran sulfate sodium,DSS)诱导的溃疡性结肠炎(UC)豚鼠的氧化应激水平的影响。方法 30只雄性Dunkin-Hartley豚鼠适应性喂养7d后,按体质量随机分为5组,每组6只:正常组(normal)[200IU/(kg·d)VD_3+100mg/(kg·d)VC]、模型组(model)[(200IU/(kg·d)VD_3+100mg/(kg·d)VC]、低D高C组(LDHC)[10IU/(kg·d)VD_3+200mg/(kg·d)VC]、低D中C组(LDMC)[10IU/(kg·d)VD_3+100mg/(kg·d)VC]、低D低C组(LDLC)[10IU/(kg·d)VD_3+10mg/(kg·d)VC],各组豚鼠采用改良灌胃法分别给予相应剂量的VD_3和VC。干预5w后,除正常组外,其余各组豚鼠自由饮用2%葡聚糖硫酸钠(DSS)溶液4d,建立UC模型,正常组自由饮用蒸馏水,各组灌胃干预不变,造模结束后,麻醉豚鼠,心脏采血,血清离心处理后于-80℃冻存待测。结果正常组豚鼠一般情况较好;与正常组相比,DSS造模后,除模型组外,其余三组豚鼠血清中1,25-(OH)_2D_3的水平升高,且LDMC组,LDLC组与模型组有显著性差别(P<0.05);与正常组相比,模型组豚鼠血清中MDA含量增高,但差异无统计学意义;VC干预组GSH-Px水平虽无统计学差异,但有降低趋势;各VC干预组的SOD水平明显降低(P<0.05)。结论 VC联合应用低剂量VD可能提高溃疡性结肠炎豚鼠血清的抗氧化水平。[营养学报,2019,41(1):53-57]
Objective To investigate the effects of different doses of vitamin C(vitamin C,VC)combined with low dose of vitamin D3(vitamin D3,VD3)on the oxidative stress level of dextran sodium sulfate induced colitis in guinea pigs.Methods Thirty male Dunkin-Hartley guinea pigs were fed adaptively for 7 days and randomly divided into 5 groups according to body weight,6 in each group:normal group(200 IU/(kg·d)VD3+100 mg/(kg·d)VC),model group[(200 IU/(kg·d)VD3+100 mg/(kg·d)VC),low D high C group(LDHC)(10 IU/(kg·d)VD3+200 mg/(kg·d)VC),low D medium C group(LDMC)(10 IU/(kg·d)VD3+100 mg/(kg·d)VC),low D low C group(LDLC)(10 IU/(kg·d)VD3+10 mg/(kg·d)VC).The guinea pigs was given a corresponding dose of VD3,VC by modified intragastric administration.After 5 weeks of intervention,the guinea pigs except those in the normal group,were free to consumeca drink containing 2%dextran sodium sulfate(DSS)4 d for the establishment of UC model.The normal group was free to drink distilled water,and the intervention for each group was not changed.At the end heart bloods were sampled,and stored at-80℃to be tested.Results The guinea pigs in the normal group were generally better.Compared with the normal group,the levels of 1,25-(OH)2 D3 were elevated in the remaining three groups of guinea pigs(P<0.05).Compared with the normal group,the MDA level in the serum of the model group was increased;but the difference was not statistically significant.The GSH-Px activity of VC intervention group was not statistically significant,but there was a decreasing trend.The SOD activity VC intervention group was significantly lowered(P<0.05).Conclusion Combination of vitamin C and low dose of vitamin D may improve antioxidant capacity in guinea pigs with ulcerative colitis.[ACTA NUTRIMENTA SINICA,2019,41(1):53-57]
