摘要
目的 探讨昆明山海棠碱诱导淋巴瘤细胞凋亡过程中caspase 3和PARP酶的作用 ,以了解THH碱诱导细胞凋亡的机制。方法 THH碱诱导JurkatT淋巴瘤细胞后 ,FAM DEVD FMK荧光标记活化的caspase 3 ,免疫细胞化学方法荧光标记PARP酶剪切后的 89× 10 3片段 ,均以流式细胞术检测。结果 THH碱能有效诱导caspase 3激活和PARP酶剪切。结论 提示在THH碱能诱导JurkatT淋巴瘤细胞凋亡过程中caspase 3和PARP酶参与调控 ,它们可能抑制了受损伤DNA的修复 。
Objective To explore the roles of caspase 3 and poly (ADP ribose) polymerase (PARP) in the apoptotic process of Jurkat T lymphoma cells induced by Tripterygium Hypoglaucum (Levl) Hutch alkaloids to understand the mechanisms of the apoptosis. Methods After induction by the alkaloids, the activated caspase 3 was fluorescenc labelled with FAM DEVD FMK and PARP cleavage was detected immunocytochemically using antibody which can recognize its 89×10 3 fragment. Then determination by flow cytometry was performed. Results The THH alkaloids could effectively induce PARP cleavage as well as activation of caspase 3. Conclusion We conclude that caspase 3 and PARP take part in the modulation of apoptosis induced by THH alkaloids, resulting in DNA fragmentation by means of inhibition of the repair of the damaged DNA.
