摘要
目的 探讨弥漫性轴索损伤后迟发性神经元损伤的病理变化。方法 以大鼠视神经牵拉损伤模型为研究对象 ,采用光镜、电镜方法对模型的损伤情况进行形态学观察 ;以TUNEL法对神经元DNA损伤情况进行观察。结果 光镜下观察见神经元数量减少和空泡化 ;电镜下观察见神经元凋亡 ,在伤后 5天最严重 ;TUNEL法见 1~ 3天陆续出现TUNEL染色阳性细胞 ,5天达到高峰 ,7天仍然存在大量阳性细胞。结论 弥漫性轴索损伤后可引起迟发性神经元死亡 ,以凋亡为主 ,持续时间较长。
Objective To explore pathological course of delayed neuronal death after diffuse axonal injury.Methods The experiments were mainly based on a rat diffuse axonal injury model. Stretch injury was induced in the optic nerves of adult Wistar rats. The conditions of tissue and cell injury were observed under microscopy and electromicroscopy. The neuronal injury was observed by TUNEL stain.Results Histological changes included the loss of neurons and vacuolation. Electromicroscopic examination showed that neuronal apoptosis occurred and reached the most serious stage 5 days after injury. TUNEL stain became positive from 1 to 3 days after injury and reached the peak after 5 days.Conclusion Diffuse axonal injury can lead to delayed neuronal death,mainly exhibiting apoptosis and lasting for longer period.
