摘要 目的 :探讨核因子κB(NF κB)在星形细胞肿瘤中的表达及其与肿瘤增殖之间的关系 .方法 :应用免疫组织化学方法检测NF κBp65 ,cyclinD1和PCNA在 64例星形细胞肿瘤组织及 10例正常大脑组织中的表达情况 .结果 :星形细胞肿瘤中NF κBp65蛋白阳性表达率为 5 4 .7% (3 5 / 64) ,cyclinD1阳性率为 4 2 .2 % (2 7/ 64) ,PCNALI(Labellingindex)为 2 .2 % - 80 .5 % ,10例正常脑组织中NF κBp65 ,cyclinD1均无 1例阳性表达 ,PCNA低表达或无表达 .NF κBp65 ,cyclinD1及PCNA的表达与星形细胞肿瘤的恶性程度有关 .NF κBp65表达与cy clinD1,PCNA的表达呈显著正相关 .结论 :NF κBp65是与星形细胞肿瘤相关的癌蛋白 ,NF κBp65上调cyclinD1,PCNA的表达 ,致使肿瘤细胞过度增殖 。 AIM: To evaluate the significance of nuclear factor kappa B (NF κB) p65 protein expression and their relationship with tumor proliferation. METHODS: The expression of NF κB p65 protein, cyclin D1 protein and proliferating cell nuclear antigen (PCNA) protein in 64 cases of astrocytic tumors and 10 cases of normal brain tissue were studied immunohistochemically. RESULTS: None of the normal tissues exhibited NF κB p65, cyclin D1 protein. In contrast, 35 of 64 astrocytic tumor specimens were positive for NF κB p65 protein with a 54.7% positive rate ( P < 0.01), and 27 of 64 astrocytic tumor specimens were positive for cyclin D1 protein with a 42.2% positive rate ( P < 0.01), the labeling index of PCNA protein were 2.2%- 80.5%. Normal brain tissue were negative or of low expression for PCNA protein. The NF κB p65 protein, cyclin D1 and PCNA protein expression were correlated to the differentiation ( P <0.05).The expression of NF κB p65 was closely related with the expression of cyclin D1 and PCNA. CONCLUSION: NF κB p65 is a new oncogenic protein related to the astrocytic tumors. NF κB p65 up regulates the expression of cyclin D1 and PCNA, then leads to tumor cell malignant proliferation and tumor occurrence.
