外周血干细胞移植患者骨髓基质细胞粘附功能的变化 被引量:9

摘要 目的 :探讨骨髓基质细胞在外周血干细胞移植 (PBSCT)预处理前后粘附功能的改变。 方法 :采用细胞粘附试验和细胞粘附阻断试验 ,观察 2 1例PBSCT患者经单用化疗 (单化 )或放疗、化疗 (放、化 )两种预处理方案后不同时相点骨髓基质细胞贴壁层对骨髓单个核细胞粘附能力的影响。 结果 :基质细胞贴壁层对骨髓单个核细胞的粘附能力 ,放、化预处理后第 30、90天显著低于移植前 (P <0 .0 1 ) ;单化预处理后第 30天显著低于移植前 (P <0 .0 5~ 0 .0 1 )。 结论 :PBSCT患者术前预处理后骨髓基质细胞粘附功能均有不同程度的降低 ,单化组的损伤较轻 ,放、化组损伤较重。预处理对骨髓微环境基质细胞粘附功能的损伤可能是影响移植后造血和免疫功能重建的重要因素之一。 Objectives:To investigate the injuring mechanism of hematopoietic microenvironment after peripheral blood stem cell transplantation. Methods:The adherent capability of bone marrow mononuclear cells and GM CFU growth on stromal cell adherence layer cultured in vitro were detected by flow cytometry, cell adhesion assay and blocking studies from patient treated with chemotherapy or chemotherapy TBI pretreatment. Results:The binding capacity of bone marrow mononuclear cells to stromal cell adherence layer from patients treated with chemotherapy TBI and chemotherapy were significantly decreased compared with that in the controls respectively( P <0.05~0.01).Contrarily the capacity of binding for chemotherapy TBI and chemotherapy patients at the time from 30 to 90 days after pretreatment were significantly decreased compared with that in the controls respectively ( P <0.05~0.01). Conclusions:The impaired adherent functions for bone marrow hematopoietic microenvironment after chemotherapy TBI pretreatment may be one of the important factors in hematopoiesis disorders of patients after peripheral blood stem cell transplantation
