基因枪法获得转基因小麦植株 被引量:12

Production of Transgenic Wheat Plants with cryIa Gene via Microprojectile Bombardment

摘要 利用适用于禾谷类作物的表达载体 pGU4ABBar,采用基因枪法 ,将人工合成的苏云金芽孢杆菌杀虫晶体蛋白基因cryIa导入小麦品种扬麦 15 8,经PCR和Southernblot鉴定 ,证明获得 4株导入cryIa基因的小麦转基因植株 ,转化率约为 0 .7% ,并通过Westernblot鉴定检测到了目的蛋白的表达。 The gramineous expression vector pGU4ABBar was used in the study. The cryIa gene, a synthetical insecticidal crystal protein gene of Bacillus thuringiensis, had been transferred into wheat variety Yangmai158 by microprojectile bombardment. The PCR and Southern blotting analysis showed that 4 transgenic plants with cryIa gene were obtained. Western blotting analysis had found the aim protein in the transgenic plants was found by western blotting analysis. The transformation frequency was about 0.7%.
